PART 1: EFFECT OF HIGH INTENSITY FOCUSED ULTRASOUND ON THE IMMUNITY OF CHILDREN WITH MALIGNANT SOLID TUMORObjective: To investigate the changes of immunity in children with malignant solid tumor treated by high intensity focused ultrasound (HIFU) by testing the changes of humoral immunity related indexes including IgG, IgA, IgE, IgM, C3 and C4 and cellular immunity related indexes including T lymphocytes conversion rate, IL-1β, IL-2 and TNF-αin peripheral blood before and after HIFU treatment.Methods: HIFU system was used to treat 20 children with malignant solid tumor. Six milliliter fasting peripheral blood was extracted from children before HIFU treatment and 7 days, 1 month after HIFU treatment. The levels of humoral immunity related indexes including IgG, IgA, IgM, C3 and C4 were tested by rate nephelometry. The levels of cellular immunity related indexes including IL-1β, IL-2, TNF-αand IgE were tested by enzyme linked immunosorbent assay. The levels of T lymphocytes conversion rate were tested by lymphocyte transformation test. ANOVA and Kurskal-Wallis Test were used to analysis the statistical of the difference among groups before and after HIFU treatment. LSD-t test were used to analysis the statistical of the difference between groups before and after HIFU treatment.Results: There were significant differences in the values of IgG and C3 between before and 1 month after HIFU treatment (P=0.015, P=0.027).No significant differences in the values of IgA, IgE, IgM and C4 were observed between before and after the ablation (P>0.05). There were significant differences in the values of IL-2 and TNF-αbetween before and 1 month after HIFU treatment (P=0.021, P=0.028). No significant differences in the values of T lymphocytes conversion rate and IL-1βwere observed between before and after the ablation (P>0.05).Conclusion: Immunity in children with malignant solid tumor treated by HIFU may be improved. PART 2: EFFECT OF HIGH INTENSITY FOCUSED ULTRASOUND ON THE DRUG RESISTANCE OF CHILDREN WITH MALIGNANT SOLID TUMORObjective: To investigate the changes of drug resistance in children with malignant solid tumor treated by high intensity focused ultrasound (HIFU) by testing the changes of the expression of multi-drug resistance gene(MDR1), multi-drug resistance-associated protein gene 1(MRP1) and lung resistance protein gene(LRP) in peripheral blood mononuclear cells before and after HIFU treatment.Methods: HIFU system was used to treat 20 children with malignant solid tumor. Two milliliter fasting peripheral blood was extracted from children before HIFU treatment and 7 days, 1 month after HIFU treatment. The levels of drug resistance related indexes including MDR1, MRP1 and LRP were tested by FQ-PCR. ANOVA and Kurskal-Wallis Test were used to analysis the statistical of the difference among groups before and after HIFU treatment. LSD-t test were used to analysis the statistical of the difference between groups before and after HIFU treatment.Results: There were significant differences in the values of MDR1, MRP1 and LRP between before and 1 week after HIFU treatment (P=0.017, P=0.014 and P=0.007).There were also significant differences in the values of MRP1 and LRP between before and 1 month after HIFU treatment (P<0.001 and P=0.006).Conclusion: Drug resistance in children with malignant solid tumor treated by HIFU may be improved.
