Objective To discuss the effect and the optimal concentration of human serum in mid-term corneal storage medium. To develop a mid-term corneal storage medium containing human serum for Chinese conditions.Methods Cornea of rabbits were stored in experimental groups A,B,C,D corneal storage medium containing human serum and control group Optisol-GS. Human serum(10%, 20%, 30%, 0%) were added into experimental groups A,B,C,D respectively. At the 3rd,7th ,10th,14th day after preservation, the viability of corneal endothelial cells was detected by trypan blue—alizarin red staining , tissue structure was detected by pathological section of cornea, the ultramicroscopic structure of corneal endothelial cells was observed by scanning electron microscope. The physicochemical property and biological safety of corneal storage medium was detected by microbiological evaluation.Results At the 14th day after preservation, there was no significant difference between group C and Optisol-GS on the viability of corneal endothelial cells, the tissue structure and the ultramicroscopic structure of cornea, P > 0.05. The physicochemical property and biological safety achieved high evaluation. At the3rd,7th,10th day after preservation, there was significant difference between group D ,A, B and Optisol-GS on the viability of corneal endothelial cells respectively, P<0.05. At the 7th day after preservation, there was no significant difference between experimental group A, B, D and Optisol-GS on the tissue structure .As time passes, the tissue structure of the experimental group D, A, B has changed in turn. At the 7th day after preservation, there was significant difference between experimental group D and groupC, Optisol-GS on the ultramicroscopic structure of cornea.Conclusion Mid-term corneal storage medium containing 30% human serum, which can preserve the corneal tissue for 7-10days, is safe and effective.
