Objective:1 To Apply Proteomics methods to screen potential markers from the bile of patients with cholangiocarcinoma.2 To investigate the expression of OLFM4 and CD146 from the bile of patients with cholangiocarcinoma.Methods:1 CleanasciteTM HC and Molecular sieveΜltrafiltration were applied for biliary proteins purification.2 The differential expression of proteins was identified by high efficiency liquid chromatography-mass spectrum (HPLC-CHIP-MS/MS).3 The results of HPLC-CHIP-MS/MS were analyzed by Spectrum Mill software.4 The OLFM4 and CD146 proteins expression was analysed by Western Blotting.Results:1 Compared with normal bile, 109 proteins were identified from the bile of patients with cholangiocarcinoma, 12 of which have not been previously detected in proteomic studies of bile. Functions of these proteins were mainly associated with cancer cellular oncogenesis, proliferation, differentiation and metastasis, etc.2 The OLFM4 and CD146 proteins expression was down-regulated in normal bile of human, but up-regulated in cholangiocarcinoma bile. Expression of CD146 was higher in the bile from lower cholangiocarcino- ma patients compared to hilar cholangiocarcinoma.Conclusions:1 Twelve differential proteins analyzed by HPLC-CHIP-MS/MS may be candidates biological molecular markers for cholangiocarcinoma.2 In cholangiocarcinoma bile, the OLFM4 and CD146 proteins expression is significantly high.
