Objective:To investigate the influence of silencing clusterin(clu) by RNA interference on proliferation and invasion of human ovarian carcinoma SKOV3 cell-line.Methods:The human ovarian carcinoma cell-line was transfected with siRNA against clu mediated by lipofectamine 2000,the expression of mRNA and protein was detected by RT-PCR and Western Blot.The proliferation and apoptosis was evaluated by MTT assay and flow cytometry ,the penetrat matrigel capacity was detected by pattern of vitro invasion .Results: The sequence--specific siRNA specifically down-regulated the expression of clu both on the mRNA and protein levels;The MTT results showed that the proliferation in experimental group was significantly slower than that in every control groups(P<0.05).The flow cytometry demonstrated that the apoptotic ratio of experimental group was significantly higher than that in every control groups,and rised about 9%,to(15.84±1.526)%(P<0.01).Compared with every control groups ,the pattern of vitro invasion demonstrated that the penetrat matrigel capacity of experimental group was significantly inhibited by clu silencing, the number of it was (26.52±6.22)cells/field(P<0.01).Conclusions:Repression of clu inhibited the capacity of proliferation and invasion on human ovarian carcinoma SKOV3 cell-line , and raised apoptotic rate obviously,the clu gene may be the new target of ovarian carcinoma treatment. Objective:To investigate the influence of silencing clusterin by RNAi on radiosensitivity of human ovarian carcinoma SKOV3 cell-line.Methods:The expression of clu gene was silenced by siRNA against it.after receive different dose of radiation(2,4, 6Gy),The MTT assay was used to detect the cell survival ratio,the cloning efficiency was evaluated by assay,and the apoptotic ratio was detected by flow cytometry.Results: The siRNA target clu specifically down regulated the expression of clu;The MTT and colony forming results showed that the cell survival and cloning efficiency was significantly lower with the dose of ionizing irradiation rise,the capacity on proliferation and cloning efficiency was inhibited obviously(P<0.05).The apoptotic ratio was significantly higher with the dose of ionizing irradiation rise,and the apoptotic ratio was to(24.32±1.75)% after 2Gy(P<0.05),raised about 12% compared with control group.Conclusions:Target to silence the clu gene can obviously enhance the radio-sensitivity of ovarian carcinoma SKOV3 cell-line in vitro,and the clu gene may be a promising target for ovarian carcinoma therapy.
