ObjectiveTransfected BDNF gene to the rat retina and visual cortex by ultrasound-mediated and estimate the protection effect on the RGC.MethodsSD male rats 88, were randomly divided into six groups, namely A normal groups (8), B sham operation groups (16), C control groups (16), D eyes transfection groups ( 16), E brain transfection groups (16), F co-transfection groups (16). After 1 week and 2 weeks of the injury, use retrograde fluorogold labeled RGC to counte the cell number, immunohistochemistry to test caspase-3, PERG to text the visual function of RGC.Results1 week and 2 weeks after the nerve injury, F Groups RGC number was 409.1±42.10 and 390.8±35.34(A Groups's 86.0% and 82.1%),significantly higher than other groups. Caspase-3 text showed that F groups at different time points, apoptotic cell number were less than other experimental groups. PERG responses for all the treated groups were normal at 1 week after injury; however, at 2 weeks, only F groups remained so(7.33±0.58μV).ConclusionUltrasound-mediated BDNF gene transfection to the rat retina and visual cortex could significantly enhance the RGC protection effect after optic Nerve Injury...
