Objective:The purpose of this experiment is to define weather HFD (Huangqi Fuzheng Decoction) have the ability to suppress the tumor growth and the expression of HIF-1αand VEGF by a model of Lewis lung cancer transplantation tumor bearing mice, The results of the subject could provide experimental evidence for the clinical application of HFD.Methods:Culture the Lewis lung cancer cell, then the Lewis lung cancer transplantation tumor bearing mice were established and randomly assigned to one of the following four groups (n=7,each group):Control group,Huangqi Fuzheng Decoction(HFD) treatment group,Cisplatin(DDP) group, Combined treatment group (HFD plus DDP). Different treatment were respectively given on the seventh day after injection subcutaneously at the same time:DDP group was treated with DDP 2mg/kg intraperitoneally injection every two days for 8 times and NS 10ml/kg intragastric administration everyday; HFD group was treated with HFD 10ml/kg intragastric administration everyday for 15 times and NS 10ml/kg intraperitoneally injection every two days for 8 times; Combined treatment group were adopted the same administration of HFD and DDP; Control group was treated with the NS 10ml/kg intragastric administration and/or intraperitoneally injection.The generral condition and body weight of mice was observed and recorded, adjust the medicine dose according to the body weight. The long diameter and short diameter of the tumor were measured every three days, then the tumor volumes were calculated and growth curves were drawn. the mice were killed on the fifteenth day after the treatment. The tumor was complete stripped and weighed, the tumor was fixed in neutral formalin and stained in the way of HE. At last we detected the expression level of HIF-1αand VEGF in Lewis lung cancer transplantation tumor with IHCResults:1. the weight of the tumors in each experimental group were lighter than those in Control group(P<0.05), the tumor inhibittion rate of HFD group,DDP group and Combined treatment group is 38.7%,52.0% and 68.96%, the weight of the tumors in Combined treatment group was lighter than other experimental groups(P<0.05),at the same time,the tumor inhibition rate is the highest in all groups(P<0.05).2. Immunohistochemistry test shows the expression of HIF-1αand VEGF decreased in each experimental group(P<0.05).Conclusion:1. Huangqi Fuzheng Decoction could inhibit the growth of the Lewis lung cancer transplantation tumor.2. Huangqi Fuzheng Decoction could increase the efficacy of DDP chemotherapy.3. Huangqi Fuzheng Decoction could inhibit the expression of HIF-1αand VEGF, this may be a mechanism of its anti-tumor effect.
