| Objectiveour previous clinical studies have proved that Fuzheng Kangai(FZKA)decoction might achieve good results in inhibiting the growth of lung cancer,prolonging survival time and improving the quality of life of the lung cancer patients,and enhancing the immune function of cancer patients.Moreover,it was demonstrated that FZKA decoction may inhibit the viability of lung cancer cells and reduce the tumor size in vivo and vitro study.However,the concrete metastasis is still unclear.So we set up this study to investigate the effects and mechanisms of FZKA decoction on the invasion and metastasis of A549 human lung cancer cells.MethodsThe inhibitory effect of FZKA decoction to the A549 cells proliferation was detected by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide(MTT)assay.Invasion and metastasis abilities of A549 cells were analyzed by Wound Healing assay.Transwell assay was performed to show the cell invasion ability by treating the FZKA decoction.Western blot assay was used to detect the expression of epithelial-mesenchymal transition(EMT)-related proteins after FZKA decoction treatment.ResultsFZKA decoction could significantly inhibit the proliferation of A549 cells in time-dependent and dose-dependent manners(P<0.05).The results of Wound Healing assay confirmed that the ability of A549 cells migration was inhibited by FZKA decoction.The inhibition on A549 cells invasiveness after FZKA decoction treatment was found from Transwell assay results.Western blot results verified that epithelial-mesenchymal transition(EMT)was inhibited by FZKA decoction in a dose-dependent mannner(P<0.05).ConclusionsThe present study demonstrated that FZKA decoction can inhibit A549 human lung cancer cells invasion and metastasis by the inhibition of EMT process. |
