Schistosomiasis japanica is a parasitic zoonosis which does great harm to human beings. Schistosoma japonicum can infect about 46 species of mammalian include human being, but either domesticated or wild Microtus fortis have natural resistance which inherits stably to Schistosoma japonicum infection. This resistance shows a new direction on research of aganist Schistosoma japonicum.In previous study, we gained a second subpool gE76 by expression cloning which had considerable anti-schistosomula effects in vitro. In order to find more gene information associated with the natural anti-schistosome effect of M. fortis, we screened second subpool gE76 by expression cloning. The chone (gE76-44) with the most significant anti-schistosomula effect was selected. Sequence alignment analysis with BLASTN in Genbank showed that it was the homologue of mouse karyopherin alpha 2 (KPNA2).In this study, we cloned Mf-KPNA2 into pLXSN, calcium phosphate transfect it into PA317 cells and packaged recombinant virus which concentrate by high speed centrifugation, with pLXSN virus and DMEM for further work after measure the virus titre.In summary, we construct pLXSN vector expressing anti-schistosome gene Mf-KPNA2, package virus and detect the activity, the virus are ready for further work.
