Objective:To investigate the effect of proliferation inhibition and radiosensitivity of Rosiglitazone on human nasopharyngeal carcinoma cell line CNE-2 and its possible sensitization mechanism.Methods:The inhibition ratio of Rosiglitazone on human nasopharyngeal carcinoma cell line CNE-2 and the mean lethal dose(IC50)were detected with MTT;The cell survival curves which embody the radiosenstivity was evaluated by cloning assay and Multi-hit single-target model;Acridine orange staining shows apoptosis of cells and flow cytometry was performed to show the number of apoptosis cells that had been staind with Annexin-V/PI;The changes of cell cycle were observed and recorded by flow cytomety which show the cells that had been stained with propidium Iodide(PI).Results:ROZ can inhibit the growth of the nasopharyngeal carcinoma cell line CNE-2 in varying degrees,and in a certain range, the inhibition ratio increased gradually with the extension of the processing time and the concentration increasing.The IC50 at 24h.48h and 72h are 114.07μmol/L,97.46μmol/L and 73.35μmol/L respectively.Cloning assay and plot survival curves showed the increase of SER and decrease of Do and Dq when CNE-2 cells were irradiated with Rosiglitazone,it definitely enhanced the radiosensitivity of CNE-2 cell line.The FCM showed that,Rosiglitazone induced apoptosis of CNE-2 cells in higher concentration,it affected obviously redistribution of cell-cycle and induced efficiently G2/M phase arrest in lower concentration.Conclusion:Rosiglitazone has the effect of proliferation inhibition and radiosensitizing effect on Nasopharyngeal carcinoma cell line CNE-2,the mechanism of radiosensitivity may be associated with blocking and redistributing cell cycle.
