Objective:We probed into the change of different fertility donor's sperm protein on the protein level, looking for several possible protein markers related with donor's sperm fertility, and new indicators for better donor screening.Materials & methods:Wu collected three kinds of donors'semen.The first kind was continuously used in 12 different recipients'AID treatment cycles, but all the women were not pregnant(low fertility); the second kind has a AID success rate in 20%-30% (normal control); the third kind has a AID success rate above 50% (high fertility).Per kind has 10 donors respectively,and per donor donated sperm 3 times.We analysed sperm motility by CASA, purified sperm by percoll density gradient centrifugation, extracted protein by RIPA lysis plus ultrasonication, detected protein concentration by BCA kit, separated total protein of donors'sperm samples by SDS-PAGE electrophoresis technique, and screened different proteins by fluorescence immunization western blotting.Results:AKAP4, GAPDHS and SPANX were differentially expressed in different fertility donor sperm, and the different protein is mostly associated with sperm motility.Conclusion:In summary, donor's fertility was associated with the sperm protein different expression, the main different protein AKAP4, GAPDHS and SPANX can be used as biomarkers for assessment of sperm fertility.Objective:We investigated the relationship between sperm acrosome reaction (AR) and the clinical pregnancy rate of intrauterine insemination (IUI), in order to guide clinical work in IUI.Materials & methods:According to certain requirements, we collected clinical data of 128 patients who accepted IUI treatment from January 2010 to December 2010 in our hospital.We purified sperm by percoll density gradient centrifugation, detectded AR by Ca2+ ionophore A23187 induction and FITC-PSA fluorescence staining, observed under a fluorescence microscope to calculate the spontaneous AR rate and the induced AR rate, and finally analysed experimental data by SPSS, to study the relationship between sperm AR rate and clinical pregnancy rate of IUI.Results:1,We compared pregnancy group with non-pregnant group, and found there was no statistical difference between their spontaneous AR rates (P>0.05), but there was significant statistical difference between their induced AR rates(P<0.05); 2,There was significant statistical difference in the clinical pregnancy rate of IUI between induced AR rate≤20% and induced AR rate 20%~50%,≥50% (4.8% vs 12.5% vs 18.6%, P<0.05).Conclusion:The spontaneous AR rate has nothing to do with the clinical pregnancy rate of IUI, but the induced AR rate is associated with the clinical pregnancy rate of IUI.
