Objective:To compare the smear layer effect to the bacteriostatic and bactericidal action of garlicin and calcium hydroxide on Enterococcus faecalis in vitro, and to Provide the evidence for the clinical use of the adequate intracanal medications.Methods:112 human posterior teeth with single root were collected after exrtaction.They were randomly assigned to 4 Experimental groups(A and B garliein groups,A and B calcium hydroxide groups) and 4 control groups(A and B control groups,A and B positive control groups)of 14 each..All of canals were prepared. The groups without smear layer were alternately flushed With 1% NaCIO solution and 17% EDTA solution, and the groups with smear layer were flushed with NS. The middle 6mm section of roots were obtained by low-speed diamond bars. The Enterococcus faecalis infection model were established. The experimental groups(56 teeth)were sealed with garlicin cotton twist and calcium hydroxide paste individually. The control groups(56 teeth)were sealed with NS cotton twist. All the teeth were placed into the anaerobic incubator.4 teeth were obtained in each group before sealing the medications,3 days after sealing the medications and 7 days after sealing the medications. The intracanal dentine power were grinded and cultured in the broth for 72 hours. The absorbance were measured.1 tooth was obtained in each group to observe the growth of Enterococcus faecalis in canal by SEMRESULTS: 1.After sealing medications 3 days,both garlicin and calcium hydroxide Inhibited enterococcus faecalis growth without Bactericidal action. Without smear layer, calcium hydroxide statistically owned greater bacteriostatic action of enterococcus faecalis than garlicin(P<0.05). With smear layer, garlicin statistically owned greater bacteriostatic action than calcium hydroxide (P<0.05)2. After sealing medications 7 days, both garlicin and calcium hydroxide owned Bactericidal action. Without smear layer, calcium hydroxide did not statistically own greater bactericidal action than garlicin(P>0.05).With smear layer, garlicin statistically owned greater bacteriostatic action than calcium hydroxide (P<0.05)Conclusion:1. After sealing medications 3 days with smear layer,neither garlicin nor calcium hydroxide could exterminate intracanal enterococcus faecalis infection completely. After sealing medications 7 days, garlicin could exterminate enterococcus faecalis effectly.2. After sealing medications 3 days,calcium hydroxide owned greater Bactericidal action of enterococcus faecalis than garlicin. After sealing medications 7 days. Both garlicin and calcium hydroxide could exterminate intracanal enterococcus faecalis infection effectly.3. With smear layer, garlicin statistically owned greater bacteriostatic action of enterococcus faecalis than calcium hydroxide.
