To Study The Relationship Between Expression Of Annexin A5 In Nasopharyngeal Carcinoma

Objective To study the relationship between expression of Annexin A5 and the differentiation and metastasis of nasopharyngeal carcinoma (NPC). And investigate the role of annexin a5 in the development and progress of NPC.Methods The expression of Annexin A5 in specimens of 65 of NPC, 18 of lymph node metastasic NPC (LMNPC) and 20 of chronic rhinitis (CR) were detected by immunohistochemistry,and the correlation between the expression of Annexin A5 and clinic factors were analysized by SPSS software. The expression of Annexin A5 in NPC cell lines with differential degree and metastasic potency were detected by western blot. Observe the change of invasive capability of NPC cells after the downregulation of Annexin A5 by siRNA.Results: 1) The positive ratio of Annexin A5 in NPC, LMNPC and CR were 84.6%(55/65), 94.4%(17/18) and 10.0%(2/20), respectively. There were significant difference in the expression of Annexin A5 between primarily NPC and LMNPC, NPC and CR (P<0.05).2) The positive ratio of Annexin A5 in moderate-differentiated NPC and poorly-differentated NPC were 64.3%(9/14) and 90.2%(46/51). There were significant difference between them (P<0.05).3) There were correlation among the expression of Annexin A5 and differentiation degree, lymph node meastasis, distant metastasis, and clinic stage (P<0.05). And without relationship to ages and gender.4) The expression of Annexin A5 in poorly-differentiated CNE2 cells were higher than in well-differentiated CNE1 cells; in high metastasic 5-8F cells higher than in low metastasic 6-10B cells (P<0.05).5) Downregulation of Annexin A5 through siRNA could decrease the invasive capability of 5-8F cells detected by transwell cave experiment. There were positive correlation betweent the expression of annexin a5 and the invasive capability of NPC. Conclusion:1) There were relationship among the expression of annexin A5 and the differentiaion and metastasis of NPC, detected the expression of annexin a5 may useful for the clinic diagnosis of NPC.2) There were relationship between the expression of annexin a5 and differentiation digree, lymph node metasatsis, distant metastasis and clinic stage.3) Down-regulation of annexin a5 could decrease in vitro invasive ability of NPC.
