ObjectiveTo explore the changes of the subsets of dendritie cells(DCs) in normal pregnant women and in the patients with unexplained early spontaneous abortion.Methods20 normal pregnant women and 21 unexplained early spontaneous abortion patients were included in this study. The DCs were stained with monoclonal antibodies against blood and decidua DCs antigens and estimated by the flow cytometric method. The numbers and subsets of DCs in the blood and decidua were Compared between the 2 groups.Results1.The numbers of DCs and myeloid DCs in the decidua was significantly higher than that in the peripheral blood(P<0.001). Conversely, the ratio of plasmacytoid DCs in the decidua were significantly lower than that in the peripheral blood(P<0.001). Compared with the peripheral blood, the ratio of MDC/PDC was also significant higher in the decidua(P<0.001).2.The level of DCs and MDC in the peripheral blood and decidua of spontaneous abortion patients are higher than normal pregnancy's (P<0.001), In the peripheral blood, the numbers of PDC was higher in spontaneous abortion patients, but not significantly(P=0.32). The MDC/PDC ratio was significantly higher in spontaneous abortion patients.3.The level of DCs and MDC in the peripheral blood and decidua of unexplained recurrent spontaneous abortion patients are higher than normal pregnancy's (P<0.001). The MDC/PDC ratio was significantly higher in URSA patients (P<0.001). In the peripheral blood, the numbers of PDC was not significantly difference(P>0.05).Conclusions1. Dendritic cells existed in the decidua. The percentage of DCs and myeloid DCs in the decidua was significantly higher than that in the peripheral blood. Conversely, the ratio of plasmacytoid DCs in the decidua was apparently lower than that in the peripheral blood. It is suggested that a focal point of function of DCs in the local environment and the role of the DCs in the peripheral blood and the decidua may be different .2. The percentage of DCs and myeloid DCs and the MDC/PDC ratio in the unexplained early spontaneous abortion patients was significantly higher than that in the normal pregnant women. it is suggested that the DCs subsets variation may be contribute to spontaneous abortion.
