Objective:To explore the frequency of dendritic cell (DC) subsets in subjects with different glucose tolerance status.Methods:The reaserch recruited221subjects with normal glucose tolerance (NGT),70subjects with impaired glucose regulation (IGR) and93patients with type2diabetes mellitus (T2DM). Clinic characteristics were recorded by study physicians. The peripheral blood mononuclear cells (PBMC) were seperated through density gradient centrifugation. The frequency of DC subsets were measured by flow cytometry. Myeloid dendritic cell (mDC) was defined as CD11c+cell, and plasmacytoid dendritic cell (pDC) was defined as CD11c-CD303+CD304+cell. We compared the frequency of DC subsets in subjects with different glucose tolerance status, and analyzed the relationships of DC subsets with obesity, hypertention, dyslipidemia, and metabolic syndrome (MetS). P<0.05was considered significant.Results:1. Compared with NGT and IGR, the frequency of pDC was significantly decreased in T2DM, while the frequency of mDC was significantly increased. There was no statistically significant difference between NGT and IGR.2. The frequency of pDC was inversely correlated with waist circumstance and was decreased in subjects with obesity, dyslipidemia or MetS; the frequency of mDC was positively correlated with BMI, waist circumstance, systolic pressure, TG, and was increased in subjects with obesity, dyslipidemia or MetS.Conclusions:Alteration of DC subsets frequency was associated with the development of T2DM.
