Objective:The aim is to investigate the association between coronary heart disease (CHD) and-12238T>C (SNP1) polymorphism and c.553G>T(SNP2)of apolipoprotein A5(ApoA5) gene and the influence of the level of serum triglyceride in Chinese Han and Uighur population of Xinjiang. Method:The polymorphism of ApoA5 gene in 486 Han patients with CHD and 501 health examination Han people 344 Uighur patients with CHD and 408 health Examination people was analyzed by polymerase chain reaction and restriction fragment length polymorphism analysis, the level of serum lipid of each patient was detected at the same time. Results:The frequencies of TT, TC and CC genotype of ApoA5 gene SNP1 were 39.92%,45.47% and 14.61% in Han CHD group while same genotypes were 33.13%,46.11% and 20.76% in Han controls group. There was significant difference in distribution of genotypes between these two groups (P=0.015). The frequencies of TT, TC and CC genotype of ApoA5 gene SNP1 were 50%, 43.31% and 6.69% in Uighur CHD group while same genotypes were 39.95%,45.1% and 14.95% in Uighur controls group. There was significant difference in distribution of genotypes between these two groups (P=0.006). There was significant difference in distribution of genotypes between Han and Uighur groups (P=0.028). Logistic regression analyses adjusted for age, gender, smoking, serum total cholesterol, presence of hypertension and diabetes revealed that individuals who carried CC genotype had an decreased risk of CHD compared with TT genotype in Han groups (OR=0.598,95%CI 0.387-0.856) and in Uighur groups (OR=0.328,95%Cl 0.154~0.700). There was also remarkably difference in level of serum triglyceride of distribution of genotypes between these two groups (P<0.05). Individuals in Han and Uighur group who carried CC and TC genotype had lower level of serum triglyceride than those carrier TT genotype. The frequencies of GG, GT and TT genotype of ApoA5 gene SNP2 were 85.39%,14.2% and 0.04% in Han CHD group while same genotypes were 91.02%,8.98% and 0% in Han controls group. There was significant difference in distribution of genotypes between these two groups (P<0.05). The frequencies of GG, GT and TT genotype of ApoA5 gene SNP2 were 93.02%,6.4% and 0.58% in Uighur CHD group while same genotypes were 94.85%,4.9% and 0.25% in Uighur controls group. There was not significant difference in distribution of genotypes between these two groups (P=0.505). There was significant difference in distribution of genotypes between Han and Uighur groups (P=0.033). Logistic regression analyses adjusted for age, gender, smoking, serum total cholesterol, presence of hypertension and diabetes revealed that individuals carried T allele(TT+GT genotype)had an increased risk of CHD compared with GG genotype. in Han groups (OR=1.756,95%CI 1.02~3.026, P=0.042). There was not significant difference in distribution of genotypes between CHD groups and controls groups in Uighur groups (P>0.05). Individuals in Han groups who carried T allele had higher level of serum triglyceride than those carried GG genotype. Individuals in Han CHD group who carried T allele had higher level of serum total cholesterol than those carried GG genotype (P= 0.014). Individuals in Uighur groups who carried T allele had higher level of serum triglyceride than those carried GG genotype(P<0.05). Conclusion:There was significant difference in distribution of genotypes of SNP1 between Han and Uighur groups. The SNP1 polymorphism of ApoA5 gene has influence on level of serum triglyceride in Han and Uighur population of Xinjiang. The SNP1 polymorphism is associated with development of CHD, and CC genotype might be a protective factor of CHD. There was significant difference in distribution of genotypes of SNP2 between Han and Uighur groups.The SNP2 polymorphism of ApoA5 gene has influence on level of serum triglyceride and total cholesterol in Han population of Xinjiang. The SNP2 polymorphism is associated with development of CHD in Han population, and T allele might be a risk factor of CHD in Han population. The SNP2 polymorphism of ApoA5 gene has influence on level of serum triglyceride in Uighur population of Xinjiang. The SNP2 polymorphism is not associated with development of CHD in Uighur population.
