Objective:To investigate polymorphism of Parkin gene V/L380 and S/N167 in Uygur and Han patients with Parkinson disease (PD) in Xinjiang,and observe whether there are difference between Uygurs and Hans. Methods:There were 168 Uygur and Han PD patients and 204 normal controls for the study, the age of 50 years as the sector was divided into early-onset PD group and late-onset PD group. The genetic DNA was extracted from all subjects.The Parkin gene's exon 10 and exon 4 were amplified by polymerase chain reaction (PCR). Then the fragments were digested by restriction endonucleases to compare the differences of the gene type and allele frequency of Parkin gene V/L380 and S/N167. Results:(1) There were no differences between Uygur and Han PD in V/L380 polymorphism allele and genotype frequency(χ2=2.409; 2.181, P>0.05); The frequencies of genetypes and alleles were significantly different between Uygur PD and Uygur control group (χ2=6.625;6.112,P<0.05);There were significant differences of the genetypes and allele frequencies between those PD group≤50 years and the control group(χ2=5.324; 4.904, P<0.05);There was a significant increase of PD in the C allele carriers (OR=2.173,P<0.05). (2) There were no differences between Uygur Ethnic PD and Han Ethnic PD in S/N 167 polymorphism alleles and genotype frequencies(χ2=2.946;2.509,P>0.05);There were no differences between PD and control in S/N 167 polymorphism alleles and genotype frequencies (χ2=1.618; 0.004,P>0.05); There were not significantly different after the comparison by ethnic, age and sex groups (all P >0.05).conclusion:The polymorphism V/L380 and S/N167 of Parkin gene were not significantly different between the Xinjiang Uygur and Han populations;The polymorphism V/L380 of Parkin gene might be associated with PD in the Uygur populations and the early-onset group;C allele might be a risk factor for onset PD; The polymorphism of Parkin gene S/N 167 might not be associated with genetic susceptibility in Parkinson disease patients from Uygur ethnic and Han ethnic,and not be associated with age and sex.
