| Objective: To determine the correlation between LRRK2gene G2385Rã€S1647Tpolymorphism and Parkinson’s disease (PD) and the differences in genotypic and allelicfrequencies between the Uygur and Han populations. Methods: Performed a case-controlstudy, which included354PD patients (171cases of Uygur and181cases of Han) and340controls (160controls of Uygur and180controls of Han).Analyzed the genotypes andallele genotypes of LRRK2gene G2385Rã€S1647T polymorphism sites by the methods ofpolymerase chainreaction (PCR-RFLP). Then verified the results by using the DNAsequencing method. Results:â‘ G2385R A statistically significant difference wasobserved between the case group and the control of Han in genotypic and allelicfrequencies,(x~2=7.873, P=0.005å’Œx~2=7.581P=0.006), Individule who carried A allelehad more risk than the non-carrier (OR=3.41,95%CI:1.42~8.19); There was also astatistically significant difference of genotypic and allelic frequencies in Han case groupcompared with the Uygur case group (x~2=16.95, P=0.000and x~2=16.95, P=0.000); TheHan individule who carried A allele had more risk than the Uygre had (OR=19.71,95%CI:4.66~83.43). According to the age group, a statistically significant difference wasobserved in genotypic and allelic frequencies in late onset group compared with theage-matched controls,(x~2=4.437, P=0.035and x~2=4.346, P=0.037), Individule who carriedA allele had more risk than non-carrier (OR=2.64,95%CI:1.07~6.50).â‘¡S1647T Astatistically significant difference was observed between the case group and the control ofHan in genotypic and allelic frequencies,(x~2=6.441, P=0.04and x~2=5.389, P=0.02).Individule who carried A allele had more risk than the non-carrier (OR=1.436,95%CI:1.058~1.950); A statistically significant difference of genotypic and allelic frequencies was also found in Han case group compared with the Uygur case group (x~2=6.127,P=0.047å’Œx~2=4.299, P=0.038); The Han individule who carried A allele had more riskthan the Uygre had (OR=1.387,95%CI:1.018~1.89).no differences were found ingenotypic and allelic frequencies between the case group and the control, and so were theUygur case group and the Uygur control, the age case groups and the age-matched control,the gender case groups and gender-matched controls is alike (P>0.05). Conclusion: Thegenotypic and allelic frequencies of LRRK2G2385R, S1647T between the Uygurs andthe Hans are obviously different; A alleles of the two polymorphism both increased therisk of PD in XinJiang Han Nationality, and have nothing to do with the occurrence ofUygur population. |
