Glioma is the most common tumor of brain, it has a higher prevalence accounting for 45% of intracranial tumors. Glioma has the character of infiltration and may be evolved from low grade to high grade tumor. Although major advances have been made in surgery and adjuvant therapy for gliomas, the mortality remains high. Recently, SNCG attract a lot of attention because it is a broad spectrum of tumor markers. Studies have shown that SNCG can be used as diagnosis markers of breast cancer, gastric cancer, colorectal cancer, lung cancer and other cancer, and have the positive correlation with the prognosis and tumor resistance.Objective:The goal of this study was to elucidate the role of SNCG in brain glioma and the relationship with the grade of tumor of glioma, and then provide a theoretical basis for diagnosis, treatment and prognosis of glioma.Methods:1. Collecting samples:43 cases of glioma samples and 10 cases normol brain tissue samples were collected from Affiliated Hospital of Medical College of Chinese People's Armed Police Force of tianjin. All patients were the first surgery for glioma, and did not receive the radiotherapy and chemotherapy before surgery.2. Samples were divided inti three groups:normal brain tissue group (N); low-grade glioma (LGG):Ⅰ-Ⅱgrade glioma; high-grade glioma (HGG):Ⅲ-Ⅳgrade glioma.3. The samples extracted the RNA quickly, and then detected the levels of SNCG mRNA in different tissues using quantitiative RT-PCR method.4. The samples were homogenized and extracted the total protein, and detected the expression of SNCG protein in different tissue using Western Blot method.5. Samples were fixed by 4% paraformaldehyde, and then dehydrated, transparent, dipped wax, embedded, sliced, maded of the paraffin sections.6. Paraffin sections were used to observe the organizational structure and determine the tumor grade of glioma by H.E stain.7. Immunohistochemistry stain was used to detected the expression of SNCG in Subcellular structures. 8. The results of RT-PCR were analyzed with gel image analysis system. The results of immunocytochemistry were analyzed semi-quantitively with the Automatic Image Analysis System Motic Med 6.0. The results of Western Blot were analyzed semi-quantitive with Image J analysis software. All the results were analyzed statistically with the software "SPSS Statistics V17.0". Statistical analysis of the results was carried out by one-way analysis of variance (ANOVA) followed by the Student-Newman-Keuls test. Differences of p<0.05 were considered significant.Results:1. The results of the RT-PCR:The level of SNCG mRNA significantly increased in LGG group and HGG group compared with normal brain tissue,2.563 times and 4.457 times to the normal group, respectively.2. The results of Western Blot:Compared with N group, the expression of SNCG significantly increased in LGG group and HGG group,2.460 times and 4.487 times to the N group, respectively.3. The results of Immunohistochemistry stain showed that in all 43 gliomas,15 cases areⅠ~Ⅱgrade glioma and 25 cases areⅢ-Ⅳgrade glioma. SNCG expression in 36 cases in all 43 cases glioma, positive rate is 83.7%, only 7 cases samples has no expression of SNCG in glioma tissues. But in 10 normal brain tissue samples have 3 cases being weak positive expression. SNCG protein majority was located in the cytoplasm as a brown-yellow or brown staining.Conclusion:The rate of SNCG positive expression is very high in brain gliomas. SNCG protein majority was located in the cytoplasm and tH.E levels of SNCG is positively correlated to the grade of glioma.
