Experimental Study On The Mechanism Of Stachyose In Treating Functional Bowel Disorder Rats Which Induced By Stomach Irritation With Cool Water

ObjectiveTo explore mechanism of Stachyose in treating Functional bowel disorder rats which induced by stomach irritation with cool water.The aim is to provide experimental evidence for clinical research.Methods40 healthy SD rats were selected and randomly divided into two groups:nor- mal group,model group.The normal group had 10 rats,they were obversed as control group,the model group had 30 rats,they were administrated by i.g. with icy water for two week. Two weeks later,selected 10 rats from model group randomly, administrated by i.p.with urethane,rats were killed to observe ultra-structure of bowel mucosa tissue.There were 30 rats after two weeks,including 10 normol rats and 20 model rats of C-IBS.The 20 molde rats of C-IBS were divided into two groups randomly: the molde conrol group,the Stachyose group.The amount of medicinal herb given to Stachyose group was 1ml/100g weight, the normal saline was given to model conrol group and normal group.The number of feces granules and the moisture content of the feces from the three groups were measured 12h after administrated by i.p.on 7th day and 14th day.Two weeks later,raised nothing except water for 16h,25 min after charcoal suspension lavaged,administrated by i.p. with urethane ,killed the 30 rats to observe following index:gastrointestinal hormones(Vasoac- tive intestinal polypeptide,Somatostatin and Motilin),small intestine charcoal sus- pension pushing rate,the number of intestinal flora,the pathology of bowel muco- sa tissue and the ultra-structure of bowel mucosa tissue.Results(1) After two weeks in threating with Stachyose,the number of feces granules, the moisture content of feces and the efficiency of the small intestine in Stachyose group was significantly increased than those in model group (P <0.05),and no significant difference compared with the normal group(P >0.05).(2)After two weeks in threating with Stachyose,the contents of VIP,SS and MOT in plasm were decreased in Stachyose group,bu it is no significant differen- ce compared with the model group(P >0.05).(3) After two weeks in threating with Stachyose,the numbers of Bifidobac- teria and Lactobacilli in Stachyose group was higher than those in model group. There were significant difference between two group(P <0.05). It is no significant difference compared with the normal group(P >0.05).(4)Observation of colonic mucosa tissue:the HE tincted microtome section of the bowels mucosa tissue of rats was not found apparente inflammatory patholo- gical changes in three groups,and there wasn't significant difference between them.(5) Observation of ultra-structure of colonic mucosa tissue:the epithelial cells and goblet cells of colonic mucosa tissue were partial denaturation in C-IBS model group.The ultra-structure of colonic mucosa tissure was significantly changed in Stachyose group.ConclusionsThe results indicated that the stachyose had the facitating feces excretion function.The mechanism of Stachyose in the treatment of FBD was to adjust the disturbance state of intestinal flora and the gastrointestinal hormones.
