| Objective:Detecting the Notch signaling pathway related protein expression level by immunohistochemistry in human giant cell tumor of bone, to explore the possible relationship between the Notch signaling pathway moleculars and the development of human giant cell tumor of bone, providing some direction and theoretical basis for finding new targets in the clinical treatment of giant cell tumor of bone.Method:Obtain 32 cases of tumor specimens and 26 cases of normal tissue specimens by surgical methods, then detecting Notch signaling protein Notch-1> Notch-2,Jagged-1 and Jagged-2 levels in tumor tissue and normal tissue by immunohistochemical staining methods,respectively. The results showed that there are some relevance between the occurrence of giant cell tumor of bone and Notch signaling pathways, so the Notch signaling pathway may make some affect on the development of giant cell tumor of bone.Results: The positive rate of the member of Notch signaling pathway Notch-1,Notch-2,Jagged-1 and Jagged-2 was 75%,62.5%,81.25% and 59.38%, in giant cell tumor of bone, respectively. When the positive rate of the member of Notch signaling pathway Notch-1Notch-2,Jagged-1 and Jagged-2 was 36.36%,22.73%,31.82% and 18.18%, in their counterpart, respectively. Making comparison between the giant cell tumor of bone and the normal tissue, the positive rate of the member of Notch signaling pathway Notch-1,Notch-2,Jagged-1 and Jagged-2 have statistical significance in giant cell tumor of bone (P<0.05).Conclusion:(1) It showed that there are some relevance of the Notch signaling pathway protein and the development of human giant cell tumor of bone.(2) Compared with the normal tissue, the expression levels of the Notch signaling protein Notch-1,Notch-2,Jagged-1 and Jagged-2 in the tumor tissue were significantly higher, which suggesting that there is a positive relevance of Notch-1,Notch-2,Jagged-1 and Jagged-2 with the development of giant cell tumor of bone. |
