Purpose:Research the expression of IFI16which is the member of interferon-inducible p200family in the bone giant cell tumors by the immunohistochemical, explore the express characteristics of IFI16in bone giant cell tumors (GCT) and clinical significanceMethods:Applicant immunohistochemical method to detect the expressions of IFI16SP proteins in51cases bone giant cell tumors of the organization of wax slice and Normal bone tissue paraffin,contrast the positive expression rate of the IFI16in the bone giant cell tumors Ⅰ-level, Ⅱ-level and Ⅲ-levelResults:IFI16protein positive expression is mainly located in the nucleus of mononuclear cells of bone giant cell tumors, the positive expression is brown particles, the positive expression is also in cytoplasm in a few cases; few positive expression in multinucleated giant cells, occasionally seen in multinucleated giant cells which is less nucleus, in this trial the total rate of IFI16protein positive expression is33.3%, and the expression of IFI16is lack in bone giant cell tumors is about66.7%, and the positive expression rate of IFI16in Ⅰ-level, Ⅱ-level and Ⅲ-level giant cell tumor of bone are64.3%,22.2%and21.1%respectively. The difference between Ⅰ-level and Ⅱ-level, Ⅲ-level is significant (P<0.05), but the difference between Ⅱ-level and Ⅲ-level is not significant (P>0.05). IFI16’s positive expression rate in1-level is significantly higher than that of in Ⅱ-level and Ⅲ-level (P<0.05). The expression of IFI16is positive in bone tissue cells of normal control group. According it, the expression of the IFI16is gradually reduced with the pathological process of the bone giant cell tumors.Conclusion:the express characteristics of IFI16:the expression of IFI16is gradually reduced along with the increase of mononuclear cells, and the gene expression may lost selectively, this suggests the possible ways are reduced transcription and improved protein degradation activities,affect the cell differentiation, play a main role in tumor progression, accord with the effect of IFI16as tumor inhibitory factor, especially in the process of cancer occurrence and development. FI16can be used as one of the auxiliary indexes in bone giant cell tumors diagnosis and pathologic stage, and has some significance in the choice of the ways of bone giant cell tumors clinical diagnosis and treatment.Bone giant cell tumors grade is higher, the more poor prognosis. Through this test, the results show that the express characteristics of IFI16is the higher the classification the lower expression, so in judging the prognosis of bone giant cell tumors, lower expression of IFI16can be used as one of the indexes...
