[Objective]To investigate the effect of EPO on expression of HSP27 and apoptosis in SD rat neurons cell with burn injury, and discuss the probable mechanism and protective of EPO for the SD rat brain tissue after burn.[Methods]We took 55 healthy SD rats (provided by Kunming medical college) to set up burn models with Standards by walker and Mason,which were randomly divided into control group,burned group and treatment group.the treatment group were administrated by injected EPO:3000u/Kg before burn injury two hour ago.meanwhile, non-treatment group were given by equivalent Physiological saline We took the brain tissue sample separately after 6,12,24,48,72 hour post burn injury. The expression of HSP27 and was detected with immunohistochemical methods and analyzed by a micro-image analysis system and the apoptosis in SD rat neurons cell was detected by tunnel, at the same time we also observed the pathological changes of the brain tissue of each groups by HE staining, and the dissolving of Nissl bodies by Nissl staining.[Results]1 HE staining Arranged neatly, cortical cell membranes of the normal group were in normal shapes and quantities. For burned group there were sparse nerve cells,widened cells gaps, and a large number of cells degeneration and necrosis which represented as inclusions shrinkage, nuclear pyknosis deeply stained and nucleolus disappeared; while the number of survival nerve cells was significantly increased andthe degree of injury wass reduced in EPO treated group.2. NissI staining Normal cortical membrane was blue-purple, light blue nucleus was full, we can see a lot of purple, and blue Nissl body, the cells arranged in neat rows; Nissl body of burn group rats were dissolved or disappearance or very few; treatment group Nissl body increased significantly, Compared with burn group.3.Immunohistochemical detection There were a little of positive expression of HSP27 in the control group, and the positive expression was observed in cytoplasm of neurons and astrocytes. In the burned group and treatment group,the expression of HSP27 was higher than the one in control group at equal and parallel time (P< 0.05),12~24h post burn injury the expression was increased, while the expression of HSP27 decreased increasedly 48h and obviously 72h post burn injury. In the treatment group, the expression was higher than the one in the burns group (P<0.05)4 TUNEL method to detect A few of apoptosis cell could be observed in neurons and astrocytes. In the burned group and treatment group, the expression of apoptosis cell was higher than the one in control group at equal and parallel time (P< 0.05), And the trend of the HSP27 consistent. However, in the treatment group, the expression of apoptosis cell was lower than burn group at equal and parallel time (P< 0.05).[Conclusion]In the early phase of severe burn injury, the brain tissue injury is very severe. It may be one of the vital mechanisms of EPO protecting brain tissue, which EPO increases the expression of HSP27 and decreases the expression of apoptosis cell in the brain tissue of SD rats in the phase of burn injury...
