Objective To explore the effects of Transforming Growth Factor-beta1(TGF-β1) message ribonucleic acid (mRNA) on repair of dogs with bile duct injury at different locations.Methods 30 Beagle dogs were randomized into three groups:controlled group, bile duct injury group and bile duct injury repair group, which contained three sub-groups:high level injury repair group, low level injury repair group, median level injury repair group. The models of bile duct injury were established and bile duct injury was repaired on varied position. Dogs were sacrificed on the 15th postoperative day and samples of bile ducts were collected. The general state of all dogs and postoperative complications were followed up. Reverse transcription polymerase chain reaction (RT-PCR) was manipulated to analyze expression of TGF-β1 mRNA.Results RT-PCR revealed that expression of TGF-β1 mRNA in bile ducts had no statistical significant in control group (P<0.05). There was remarkable difference relate to TGF-β1 mRNA at the three levels among BDI group (P<0.05). In BDI-repair group, TGF-β1 mRNA of high level was lower than that in median level or low level (P<0.05). But between the low level and median level, difference of TGF-β1 mRNA had no statistical significance (P> 0.05). At low level, expression of TGF-β1 mRNA in controlled group was remarkably lower than the corresponding level in BDI-repair group and BDI group (P<0.05),but differences between in BDI-repair group and BDI group had no statistical significance(P>0.05); At the median level or high level, expression of TGF-β1 mRNA, which in the BDI-repair group, was significantly different when it compare to corresponding indicator of BDI group and control group (P<0.05),but difference between controlled group and BDI group had no statistical significance (P> 0.05).Conclusions:The quantity of expression of TGF-β1mRNA in bile ducts of injury repair increased, and increments were various on different levels. TGF-β1mRNA expression was relatively higher at sites nearly to duodenum (low level) than that at sites near to hepatic portal (high level). Under same condition of BDI, expressions of TGF-BjmRNA were higher when samples were derived from the lower level in BDI-repair group. Because of scar-related factors (TGF-β1mRNA) expression after repair and postoperative incidence of restenosis of bile duct were highly correlated, the incidence of restenosis was lower when the bile duct-jejunum anastomotic level escalated.
