Objective:To prepare Lewis lung cancer cells/dendritic cells fusion vaccine (HC) and rsearch its biological characters, anti-tumor effects.Methods:Bone marrow separated from mice, were co-cultured with cytokines to generate mature DCs. The Lewis lung cancer cell lines were fused with DCs by polyethylene glycol (PEG). Hybrids (HCs) were administered by four strategies:protective immunization, vaccination, adoptive cellular therapy, and vaccination combine adoptive cellular therapy.Results:The Lewis lung cancer cells/dendritic cells fusion vaccine (HC) were prepare successfully. In protective immunization, HCs induced significant retardation of tumor growth. Similar results were observed in vaccination HCs induced activation of proliferating cytotoxic T cells and a significant retardation of tumor growth. When T cells from HC-vaccinated mice were transferred into naive tumor-bearing mice, tumor growth was retarded.An efficient proliferation and cytotoxic T-cell response was observed. But the effects of adoptive cellular therapy were inferior to vaccination. Finally, the effects of vaccination combineing adoptive cellular therapy were inferior to vaccination Conclusions:Dendritic cells/Lewis lung cancer cell line fusion vaccine (HC) offer active anti-tumor effects to mice lung cancer.
