Objiective:To compare the cytotoxicity of low-osmolar contrast media, iso-osmolar contrast media and Na+,K+-ATPase activity of human proximal renal tubular epithelial cell (HK-2),and to explore the difference of acute kidney injury between different osmolar and different dose, then to investigate the pathogenesis of contrast-media induced nephropathy (CIN)Methods:HK-2 grow in normal conditions, divided them into culture media control and contrast-media treated groups with low-osmolar contrast media and iso-osmolar contrast media.HK-2 were exposed to culture media and contrast media with 3 different dose (30ul/ml,60ul/ml,90ul/ml) for 24h,48h and 72h.The cytotoxicity of HK-2 was assessed by MTT assay.The activity of Na+,K+-ATPase was detected by shadepilot with ATP kit.Results:Compared with control group, contrast-media severely increased the cytotoxicity of HK-2 (P<0.05, P<0.05),and iso-osmolar contrast media is better than low-osmolar contrast media (P<0.05).During the study time, the cytotoxicity increased in a time-dependent and dose-dependent manner(P<0.05, P<0.05).Compared with control group, Na+,K+-ATPase activity of HK-2 was inhibited significantly in contrast-media group (P<0.05), and iso-osmolar contrast media is better than low-osmolar contrast media (P<0.05). The activity of Na+,K+-ATPase decreased in a time-dependent and dose-dependent manner, and there's significant difference in the time 24h,48h,72h (P<0.05).There's significant difference between the three doses (P<0.05). The activity of Na+,K+-ATPase of HK-2 inverse correlated with the cytotoxicity of HK-2(r=-0.794, p<0.05).Conclusion:Contrast-media severely increased the cytotoxicity of HK-2, and also decreased the activity of Na+,K+-ATPase of HK-2 significantly. Compared low-osmolar contrast media with iso-osmolar contrast-media, iso-osmolar contrast-media is better than another one. The contrast dose is one of the factors which may induced nephropathy. The activity of Na+,K+-ATPase of HK-2 inverse correlated with the cytotoxicity of HK-2.
