| ObjectiveThis study through the basic research and clinical study, with endothelial markers as the standards of evaluation of endothelial function, to study the improvement action of tong guan capsule on atherosclerotic vascular endothelial function.Methods1. Basical ResearchThirty-six New Zealand rabbits were randomly divided into six groups, including the blank control group, the high lipid control group, the atorvastatin group, the large, medium and small dose of tong guan capsule group, the blank control group feeding basic food, the high lipid control group feeding high-fat food, the atorvastatin group adds atorvastatin (5mg/kg·d) to high-fat food daily, the large, medium and small dose tong guan capsule groups respectively adds 15mg,9mg and 3mg tong guan capsule to high-fat food daily. We collect TC, TG, HDL-C, LDL-C, NO, ET and vWF at the second weekend, forth weekend and sixth weekend and at the sixth weekend we do the pathology testing with iliac artery. spss16.0 was used to do statistical analysis, all analysis use bilateral test, P<0.05 was statistically significant.2.Clinical ResearchEighty-seven clinic patients who were diagnosed stable angina and with stagnation of heart blood or stagnation of phlegm or qi stagnancy of TCM syndrome differentiations were collected, all the cases were randomly divided into the tong guan capsule group, the atorvastatin group, the tong guan and atorvastatin group, the tong guan capsule group patients take the tong guan capsule (3 capsules,'tid) for 3 months, the atorvastatin group patients take atorvastatin (20mg, qn) for 3 months, the tong guan capsule and atorvastatin group take the tong guan capsule (3 capsules, tid) and atorvastatin (20mg, qn) for 3 months, we collected blood lipid indices, vWF, NO and ET before and after the study. At the beginning of the study the patients were divided into stagnation of heart blood group and without stagnation of heart blood group for the statistical testing, spss16.0 was used to do statistical analysis, all analysis use bilateral test, P<0.05 was statistically significant.Results1.Basical Research1. The TC of the large-dose tong guan capsule group significantly reduced with increasing HDL, the difference was statistically significant (P<0.05)2. There was no statistical significance(p>0.05) of the vWF and the ET level for the large-dose tong guan capsule group and the atorvastatin group before-and-after the study, but there was significant increasing with the NO level (p<0.05).3. The histopathological results showed that the control group, the atorvastatin group and the large-dose group arterial lining were smooth, there was no plaque formation, other groups have different degree of arterial lining plaque formation, the medium and small dose groups also controlled the formation of the plaque at a certain degree compared with the high-fat group, but the effect obvious weak.2. Clinical Research1. There was no statistical significance for the distribution of the TCM syndrome differentiations among the tong guan capsule group, the atorvastatin group, the tong guan capsule and atorvastatin group.2. There was no significance of the vWF and NO level between the stagnation of heart blood group and without stagnation of heart blood group at the beginning of the study, but the ET level of the stagnation of heart blood group was higher than that of the other(p<0.05).3. The ET level of the tong guan capsule and lipitor group was higher than the other two groups before the study, the difference was statistically significant (p<0.05), and there was no difference of the ET of the three groups after the study (p>0.05);there was no significant difference of the NO and vWF level of the three groups before the study (p>0.05), the NO level of the tong guan capsule and lipitor group was higher than the other two groups, but the vWF level was lower, all the differences were statistically significant (p<0.05).ConclusionThe tong guan capsule reduces the level of blood TC, TG, LDL-C, and increase the level of the blood HDL-C, at the same time reduces the blood vWF and ET level of the rabbit and the patient, but improves the level of NO, so it can greatly improves endothelial function of the patients with coronary disease; we also found that the stagnation of heart blood group take larger percentage than the other in the patients with coronary disease,and the ET level of the heart blood group is higher than the other... |
