Transdermal drug delivery system (TDDS) attracts more and more attention for its advantages. However, the skin barrier limits the development of TDDS.Chemical enhancers and physical methods have been used for enhancing transdermal drug transportation.In order to develop a newly transdermal delivery system and channel cataplasm and then to promot the development of Chinese medicine, this thesis focused its attantion on the preparation procedure, the transdermal permeation property in vitro and the quality evaluation.The research mainly includes the following aspects:1,Study on the preparation procedure of Channel CataplasmA appropriate blank matrix is screened out and the bigger influential, factors of matrix are preliminarily determined through the preliminary test and single-factor experiment.The L9(34) orthogonal test method is used and tacking strengthcohesive strength and physical properties are used as evaluation indexes.The optimal matrix prescription of channel cataplasm is Viscomate NP-700:6.0g, Dihydroxy alumimun aminoacetate:0.14g, Carbomer 940:2.0g, PVP:4.0g,Glycerol:36g, Tartaric acid:0.20g, EDTA:0.05g, Water:70.0g, ultramicro power of Ginseng and Panax Notoginseng:10g, Ginger oil:0.6mL.With this optimal matrix prescription better quality cataplasm can be prepared.2,Study on the transdermal permeation in vitro of Channel CataplasmThe experiments of transdermal permeation in vitro are conducted by using Franz diffusion cell.With the amount of accumulation permeation (Q) as index, all of the permeation enhancers is reseached, and the optimized combination are 3% Azone,1% OleieAeid1,3% propylene glycol. The amount of 24h accumulation permeation is obtained as 701.076±17.739μg·cm-2, and the permeation rate is 26.317μg·cm-2·h-1.3,Quality evaluation researches on Channel CataplasmOn the research of the quality standard, the weight differences and shaping meet Pharmacopoeia of the People's Republic of China of 2010 edition. By the adhesion experiment, the result shows that it can stick on the 21th ball (weight 12.0g), and cohesive strength time is 80s. Ginger oil is qualitatively tested by TLC, which shows a good resolution and specificity. The content of notoginsenoside R1, ginsenoside Rgl, ginsenosid Rb1,6-gingerol in Channel Cataplasm are determined by HPLC.The amout of four components is 46.569mg per Cataplasm.
