ObjectivesThrough our animal experiments, we aim to prove that the cause of functional dyspepsia is closely related with gastrointestinal motility disturbance as well as parasecretion of gastrointestinal hormone like motilin and somatostain. Meanwhile we want to discuss the question that whether Simo decoction regulates gastrointestinal motility by its affect on gastrointestinal hormone levels such as motilin and somatostain. Understanding the pathophysiological course of functional dyspepsia, helps us to study the possible pathway and mechanism of Simo decoction on regulating gastrointestinal dysfunction. Furthermore, we can also provide more modern science evidences and theoretical support for using Chinese herbs to treat functional gastrointestinal diseases.Methods1. Fifty rats were irritated by clamping tails for making FD models. Then the rats were divided randomly into 5 groups:high, medium and low dosages of Simo decoction, Mosapride tablets, and model group. At the same time, we chose ten healthy rats as the blank group with equal number of male and female rats.2. After a treatment for one weeks, the content of motilin and somatostatin in plasma of Simo decoction different dosages groups were examined through the radioimmunoassay method, and gastric emptying was also observed. Then the outcomes were compared with that of model group and Mosapride tablets group.Results1. Compared with that in the blank group, the content of motilin in model group significantly decreased (P< 0.05), while the gastric emptying and the content of somatostatin obviously increased (P< 0.05).2. Compared with that in the model group, the content of motilin in the Simo decoction high dosage group, medium dosage group, and the Mosapride tablets group obviously increased (P< 0.05). Meanwhile the gastric emptying and the content of somatostatin significantly decreased(P< 0.05).Conclusions1. Parasecretion of motilin and somatostain can lead to gastrointestinal motility dysfunction. Our findings demonstrate that motilin and somatostain secretion disorder is one of the pathogenic factors of functional dyspepsia.2. Simo decoction is an effective Chinese medicine for treating functional dyspepsia.3. Simo decoction promots gastric emptying in functional dyspepsia model rats. The mechanism might be related to its adjusting the release of motilin and somatostatin, and its influence on the release of Vagus Nerve's neurotransmitters.
