BackgroundCerebral vascular disease is regarded as one of the three major causes of death. Separately, the incidence, morbidity and death rate are very hight. About 50% to 70% of them left sequelas, such as hemiplegia, anepia, dementia, etc. Nerve cell necrosis and the reperfusion injury result in the neurologic impairment and hemiplegia after cerebral ischemia. Recently research indicated, The neuranal repair after cerebral ischemia is not only concerned with nutrion factor, growth factor and blood vessel neogenesis, but also closely correlated with CNS inhibitive factor to impede nerve regeneration. The effect of inhibition factors prevent nerve regeneration of ACT. Accordingly they are the cause made sequelas, such as hemiplegia or aphasia. Recently discovery that Nogo-A is the most fervent nerve fiber regeneration inhibitive factors. It combines with the receptor NgR to prevent nerse regeneration. This is the important cause of sequelas. It seems that NgR is the focus to producea marker effect of some axon growth repressible proteins in CNS myelin. This offers new direction for CNS regeneration and repairation study.It's considered to control NgR could promote the injury CNS nerve cells functional recovery greatly and reverse the bad consequence.So to reprenss conduction of inhibitive factor cold improve the CNS function. Some research indicated that electric acupuncture treatment is a modus operandi to promote ACI nerve functional recovery.But there is not any report about whether the mechanism relate to the inhibition of NgR expression both internal or abroad. Therefor it's a new way to prove if electric acupuncture treatment can promote CNS nerve functional recovery and what is the mechanism. That will be helpful to interpret what is the mechanism it do to CNS nerve, and provid a new method for clinical application.Methods130 male Sprague Dawley rats (240+40g) were randomly divided into electro-acupuncture group(30 rats), sham acupoint group(30 rats), model group(30 rats), sham operation group(30 rats) and blank group(10 rats). The first three groups were adopted modified method of ZeaLonga to prepare The left middle cerebral artery occlusion(MCAO). After the operation of model group and electro-acupuncture group were selected "Baihui"(DU20)and "Dazhui"(DU14) points were given electro-acupuncture treatment everyday. After sham acupoint group was selected the sides of "Baihui"(DU20)and "Dazhui"(DU14)points 1 inch there were given electro-acupuncture treatment everyday. The sham operation group was only operated operation wound. Using The immunohistochemistry and The transmission electron microscopy to observe electro-acupuncture after cerebral ischemia 2hs-reperfusion 1,7,28day, the expression of bilateral Cerebral Cortex NgR of each group rat and effect of Ultrastructureonset and compare with the model group and the sham electro-acupuncture group.Results1,Every mouse of model group, electro-acupuncture group and sham acupoint group had significant neurologic impairment after ischemical reperfusion injury 1 and 7 duys later. And the nerve function scores were patency different to sham operation group. Neurologic impairment of electro-acupuncture group reduced more than model group or sham acupoint group, but the same with sham operation group 28 days later.2,Neurologic impairment of model group, electro-acupuncture group and sham acupoint group had significant recovery after ischemical reperfusion injury. And the recovery of electro-ocupuncture group was the most rapidly, or sham ocupoint group was more slowly than electro-ocupuncture group.3,The NgR mase cell population in the damage cerebral cortes of model group, electro-acupuncture group and sham acupoint group rise afer ischemical reperfusion injury 1 days later. The maximum happened 7 days after ischemical reperfusion injury, and it decresed 28 days. The NgR mase cell population of electro-ocupuncture group was less than other groups from 1 to 28 days later. The populations of electro-ocupuncture group were the same with sham operation group 28 days later.4,The NgR mase cell population in the uninjured side cerebral cortes of model group, electro-acupuncture group and sham acupoint group rise at 1,7 and 28 day after ischemical reperfusion injury. The NgR mase cell population of electro-ocupuncture group decreased better than other groups. The population of model group, electro-acupuncture group and sham acupoint group was the same 28 days later.ConclusionsThe NgR expression rise after ischemical reperfusion injure both the damage cerebral cortes and the other. Electric acupuncture treatment on Baihui and Dazhui could promote neurologic impairment recovery and down regulate the NgR expression to protect the injury nerve cells.
