ObjectiveAcne is a kind of common and easy onset skin diseases, and it is a associated product of dolescence. TCM theory think that Acne owing to the heat from Lung, Spleen, or stomach. That the heat together with pathogenic factor result in blood-heat sluggish in the skin. With the advance of the human civilization, and the improvement of living standard, people pay more and more attention to the quality of sexal functional cosmetics, and natural herbs acne removing cosmetic begin more and more loved by people. The prescription which are consist of Radix scutellariaen Cortex phellodendri,Radlx sophorae flavescentis,Radix et rhizoma rheit(the proportion of them is each kind occupying 1/4), is proved to be efficient through clinical verification. Among the four kind of traditional Chinese medicine, adix scutellariae can remove the heat and the moisture of The Body away;, and it role in the lungs; Cortex phellodendri can also remove the heat away, and it role in the lower part of humen bodies;Radix sophorae flavescentis always used for a variety of skin diseases; Radix et rhizoma rhei can also have some effect of clearing heat and accelerating blood circulation. In recent years, The industry of Traditional Chinese Medicine cosmetics develops rapidly, while the technology of the whole vocation which include prescription,craft,quality,and efficiency, in especial the extraction and separation technology of TCM cosmetics, are backward. This study uses advanced technology and syatematically research, so that the active component of the prescription can get retained greatly and the final procuct of the prescription can contains higher levels of active constituent, and its colore light and brigh. At the same time, provide demonstration for the craft of extraction and purification of TCM compound which are used in cosmetics.MethodsResponse surface methodology was used for optimizing the extraction of the TCM compound for removing acne at the first time. Through preliminary experiment and a lot of literature, certify that using ethanol for extraction solvent have higher remaining rate than using water. In this study ethanol concentration (A), ethanol consumption (B), extracting time(C) were used for the three investigating factors, and each of them designed three levels. HPLC method was for determining the transferring rate of emodin, rhubarb phenolic, baicalin, and berberine, which were the index of the compound. Analysis the significantion and interactions of the three factors. The predicted values of the methd was proved by experiment. At last, determined the best extraction parameters.In this study Macroporous resin process with single factor experiment was used for purification at the first time. HPLC methd and TLC method were used for determining the transfering rate of emodin, rhubarb phenolic, and baicalin, Berberine. HPLC methd and TLC method were used for determining and calculateing the transfering rate of active ingredient. static adsorption experiment were used for selecting the four types of Macroporous resin, as, D101, compound—JD, AB—8, HPD750. Using dynamic resolution to investigate the concentration and dosage of the ethanol that were used for elution concentration.ResultsThe optimal parameters for TCM compound extraction were:ethanol concentration for 75%, ethanol consumption for ten times of the weight of the compound recipe;extracting time for half and two hours at the first time (fixed ethanol consumption for eight times of the weight of the compound recipe and extracting time for one hour in the second time). The three batches of validation experiment draw that the average transfering rate of hubarb phenolic is 70.71%, of emodin is 52.89%, to baicalin is 79.62%, and for berberine is comes to 84.69%. Those are close to the Prediction by the model of a equation for the extraction rate of the mostly effective ingredient. Certificating that the mode is dependable. The extraction under the optimum conditions was enrichment to have no flavor of Alcohol, thinking about the solubility of Anthraquinone class, concentrated liquor was added a certain proportion of ethanol. through reviewing and analysising, get that when liquor include ethanol 20%, the transfer rate of effective ingredients effects is highest.Screen out the macroporous resin D101 as the optimal macroporous resin, from the macroporous resin D101, compound—JD, AB—8, and HPD750. It testify that the choosed macroporous resin have both good adsorption and analytic effect.For the concentration of eluent, designed a concentration gradient of 55%,65%,70%,75%,85%,95%, proved that 65%ethanol can do best to the elution of baicalin. But 65%ethanol is inefficient for rhubarb phenolic as the eluan. To rhubarb phenolic, the sparation effcient is becoming better and better as the concentration of ethanol increased. So fist used 65% ethanol, and the 95%ethanol for eluting.The end of elution adopted method TLC. It result in that when using 65 % ethanol to elute baicalin. When elution comes to 10times of the volume of the macroporous resin D101, there are hardly spots of darker color. And there are no spots of hubarb phenolic, when it used 65% ethanol to elute. Instead of it, using95% ethanol, there are spots of hubarb phenolic, and when its consumption comes to the 6th volume of the resin, the spots of hubarb phenolic are very weak. Both 65% ethanol and 95% ethanol can elute berberine, and when the volume of elution liquid comes to the 5th volume of the resin, the spots of berberine disappear.Though the optimization,get a packaged artwork, as ethanol elxtraction——macroporous resin separation.conclusionThrough response surface methodology, The optimal parameters for TCM compound extraction were:alcohol concentration for 75%, Alcohol consumption for ten times of weight a, extracting time for half and two hours at the first time(fixed ethanol consumption for eight times of the weight of the compound recipe and extracting time for one hour in the second time). The result of the three batches of validation experiment are close to the Prediction by the model of a equation for the extraction rate of the four indicators, which proved it to be high reliability, when liquor include 20% ethanol, the transfer of the mostly effective ingredients are best. Of the four kinds of Macroporous resin, resin D101 ows high efficiently and low price. The elution condition was:first 65% ethanol for 10times of the volume of resin, and then 95% ethanol for 6 times of the volume of resin.
