Paliurus ramosissimus(Lour.)Poir,the main chemical composition is triterpenes,total flavonoids and coumarins and so on,the pharmacological effects of the treatment of sore carbuncle ulcers,detoxification swelling.In this study,the chemical composition of betulinic acid and paliurusene were excellent antitumor activity.Taking the two compounds as the index components,First study the single factor experiment,and then the L9(3~4)orthogonal experiment was used to optimize the extraction process.Finally,the optimum extraction process was as follows:After the fresh leaves,5 times the amount of 95%ethanol,at pH 2 under reflux conditions for 80min,the residue was continuously refluxed at pH 8 for 40min,combined the extract,cooled and filtered,and dried to the dryness.The results show that the process has good stability and high reproducibility.By comparing the other extraction methods,the semi-bionic method is superior to the cold-maceration,which is superior to the Ultrasonic Extraction.By comparing the static adsorption and dynamic adsorption of paliurusene and betulinic acid with different macroporous adsorption resins,AB-8 macroporous resin was selected as the best resin for the purification of Paliurus ramosissimus,and the adsorption and elution process were optimized.The best purification process parameters were as follows:the concentration of paliurusene was 16.07?g/mL,the concentration of betulinic acid was 1023?g/mL,the loading speed was 3mL/min,the better amount was 0.4g of the amount of alcohol/g dry resin can achieve a better adsorption effect,the adsorption rates were 87.1%and 82.03%,respectively.85%ethanol as eluant,the amount of 900ml,elution rate of 3mL/min when the two active ingredients can be eluted,the elution rates were 88.23%and 73.52%.In this effective component,the content of paliurusene was increased by 8 times compared with that of the extract of ethanol extract,and the content of betulinic acidwas increased by 5times.At the same time,the purification process removed a large number of pigment components,so that the effective components of the traits have been improved.To explore the anti-tumor material base of Paliurus ramosissimus,the in vitro antitumor tests of ethanol extract,ethyl acetate extract and effective component were studied respectively.And the alcohol extracts were separated and purified to obtain 6monomer compounds,Hela,MTT was used to detect the antitumor activity.The results showed that paliurusene,paliurusene A,paliurusene B,messagenic acid B have good antitumor effect.The antitumor activity of the double substituents is stronger than that of the monosubstituted compounds,and the single substitution is stronger than that of the mother nucleus.The quality standard of virgin of the Paliurus ramosissimus was established.And bivalve acid content of not less than 0.7%and 40%.Three batches of effective components of the Paliurus ramosissimus were tested for accelerated test and long-term stability test.The results showed that the inspection are in line with the provisions.
