Partâ… :Effects of ginsenoside-Rd on the expression levels of netrin-1 and neuron apoptosis in pilocarpine induced epilepsy model.Objective:To investigate the effects of ginsenoside-Rd on the expression levels of netrin-1 and neuron apoptosis in the rat hippocampus after lithium-pilocarpine induced epilepsy. Methods:Thirty Wistar rats were randomly enrolled into the NS control group (NS), temporal lobe epilepsy group(TLE) and ginsenoside-Rd group (GSRd). The epilepsy animal model was established using lithium-pilocarpine. netrin-1 proteins were detected with immunohistochemistry method. The neuron apoptosis was observed with TdT-mediated dUTP nick end labeling(TUNEL). Results: The expression of netrin-1 in GSRd group was notably lower than that of TLE group(p<0.05). TUNEL positive cells in TLE group significantly higher than that of NS group (p<0.05). TUNEL positive cells in ginsenoside-Rd treated group significantly decreased as compared with TLE group (p<0.05). Conclusion: Ginsenoside-Rd may serve as an effective agent for curing the brain damage after TLE in vivo; the mechanism of protecting the neuron may include downregulating the expression of netrin-1, reducing the number of apoptotic neurons.Partâ…¡:Expression of netrin-1 in the process of mossy fiber sprouting after pilocarpine-induced epilepsy model.Objective:To explore the correlationship between axon guiding cues netrin-1 and the process of aberrant mossy fiber sprouting (MFS) following status epilepticus (SE). Methods:The temporal lobe epilepsy animal model was established using lithium-pilocarpine. The process of aberrant mossy fiber sprouting and the expression of netrin-1 was observed with Timm and immunohistochemical staining. Results:At 2 and 4 weeks after seizure,netrin-1 immunostating showed an increase of netrin-1 in the dentate gyrus and MFS was found after temporal lobe epilepsy. Conclusion: netrin-1 may be involved in the process of mossy fiber sprouting.
