Objective Many studies have shown that carbamylated erythropoietin (CEPO) has clearly cardio-protective effect, but lacks the report of chronic heart failure (CHF). We investigated the effects of CEPO on hemodynamic parameters, the serum level of matrix metalloproteinase-2 (MMP-2) and tissue inhibitor of metalloproteinase-1 (TIMP-1), left ventricular mass index (LVM1) and the volume fraction of collagen (CVF) to determine whether CEPO has cardio-protective effect in rats of CHF and the possible mechanisms.Method 10 of 80 healthy adult Wistar rats have been randomly selected as control group, others were injected isoprinosine (ISO),5mg/kg, once a day, continued three days, establishing the model of CHF. Five weeks later, monitoring the hemodynamic parameters, we judged the succeed standard by left ventricular end diastolic pressure (LVEDP)≥15mmHg, then rats were divided into CHF group and CEPO group. CEPO group were intraperitonealy injected CEPO 50μg/kg, twice a week, last four weeks, while CHF and control group were receiving the same volume of normal saline at the same time. After four weeks, hemodynamic parameters, LVMI, the level of serum MMP-2 and TIMP-1, CVF were evaluated.Result Compared with control group, left ventricular systolic pressure (LVSP), maximal rate of rise/decline of left ventricular pressure (±dp/dtmax) (P<0.01 for each) were significantly decreased, left ventricular diastolic pressure (LVDP) and left ventricular end diastolic pressure (LVEDP) were significantly increased (P<0.01), the level of serum MMP-2, LVMI and CVF were higher, but TIMP-1 was lower (P<0.01) in CHF group; compared with CHF group, CEPO group have higher LVSP,±dp/dtmax and the level of serum TIMP-1 (P<0.01), lower LVDP, LVEDP, the level of serum MMP-2, LVMI and CVF (P<0.01).Conclusion CEPO can reverse myocardial remodeling, improve cardiac function in rats with CHF by regulating the expression of MMP-2, TIMP-1 and degradation the volume of collagen.
