Objective:1,Through the immune injection method to establish SD rats models of recurrent aphthous ulcer (RAU). It could provide an occurrence and therapy background for human beings recurrent aphthous ulcer.2,To explore the effects of Lycium barbarum polysaccharide (LBP) on experimental RAU model in SD rats and the influence on its immune mechanism.Methods:1,80 rats were randomly divided into 2 groups:normal control group (16) and experimental group(40),24 is extracted oral mucosa as antigen.2,The mixstrue of oral mucosa extracted from 24 SD rats and complete Freund's adjuvant were applicated to immune rats as antigen. Antigen above added adjuvant was injected to intraderm adjacent the SD rats spine once a week and lasted for 8 weeks to observe the occurrence and cicatrization of oral ulcer in the experimental rats. The negative control group given 0.1 ml NS (normal saline).3,After 8 weeks, histopathology examination was performed with oral mucosa both two groups.4,The expression EGFR in oral mucosa was measured by immunohisto chemical.5,The expression EGFR in SD rats serum was examined with sensitive enzyme-linked immunosorbent assay(ELISA) before and after establishment of RAU rats model.6,The values of CD4+, CD8+ cells and CD4+/CD8+ in SD rats of each group were measured by flow cytometry (FCM) after modle.7,40 rats were randomly divided into 5 groups:normal control group (A),negative control group (B),100mg/kg LBP treatment group (C),200mg/kg LBP treatment group (D),300mg/kg LBP treatment group (E).8,After 20 days' administration, the histopathology examination was performed with oral ulcer mucosa.9,The expression EGFR in SD rats serum was examined with sensitive enzyme-linked immunosorbent assay (ELISA) after 20 days'administration.10,After 20 days'administration, the values of CD4+, CD8+ cells and CD4+/CD8+ in SD rats of each group were measured by flow cytometry (FCM).11,2 days' after administration, explore the effect of LBP on thymus quality and thymus index of rats in different groups.Results:1,The expression of EGFR in SD rats serum in control group was 833.79±7.41 and 837.43±4.58 respectively (P>0.05),in experimental group before and after modal was 843.54±3.87 and 282.69±7.91 respectively (P<0.05).Compared with control group, the expression of EGFR in experimental group was deceased, the difference between two groups was significant (P< 0.05).2,Immunochemical analysis of EGFR demonstrated both in normal and RAU mucosa, and the positive rate of EGFR in normal and RAU mocusa was 30.7±10.1 and 9.6±6.2 respectively,the difference between two groups was significant (P<0.05).3,After modle, the ratio of CD4+,CD8+ cells and CD4+/CD8+ in normal group was 49.91±3.27,18.72±2.45,2.45±0.32 respectively, in experimental group was 30.27±3.53,29.76±4.02,1.01±0.78 respectively. Compared with normal control group, the ratio of CD4+ and CD4+/CD8+ was significantly decreased, while CD8+ obviously was increased in experimental group, the difference was significant (P<0.05).4,After 20 days' LBP administration, in five groups, the expression EGFR expression in serum in normal control group(A) was 824.61±4.36,in experimental group(B) was 303.25±7.81, LBP(100mg/kg)(C) was 492.31±3.24,LBP(200mg/kg)(D) was 492.31±3.24, LBP(300mg/kg) (E) was 804.26±4.60.The EGFR expression in SD rats in serum of negative control group(B group) were significantly lower than normal control group(A) (P<0.05).Compared with negative control group, the EGFR expression in SD rats in serum of LBP groups was increased,the difference was significant (P<0.05).Furthermore,the EGFR expression in LBP(300mg/kg) (E) was greater than other two groups.5,After 20 days' LBP administration,compared with normal group,the ratio of CD4+ and CD4+/CD8 was decreased, the ratio of CD8+ was increased,there is a significant difference between negative control group and normal control group (P<0.05).Compared wihe negative control group,the ratio of CD4+,CD4+/CD8+rose in rats of LBP treatment groups and CD8+ was down (P<0.05).6,The thymus quality and thymus index in negative control group was 314.6±78.2 and 73.3±16.9 respectively, and lower than in control group,the difference was significant (P<0.05). The thymus quality and thymus index in LBP treatment groups was higher than negative control group, the difference was significant (P<0.05).Conclusion:1,Oral mucosa was used as antigens to immunize SD rats. The ulcer was very similar to RAU in human beings clinically and pathologically. Our preliminary results showed that this animal model could be used as model of RAU of human beings for further study on its pathogensis.2,The expression of EGFR both in normal oral mucosa and RAU oral mucosa,and decreased in latter.This may be related to the mechanisms involved in RAU formation.3,The ratio of CD4+ and CD4+/CD8+ significantly decreased in T-lymphocyte is related to the pathogenesis of RAU.4,LBP could increase the counts of CD4+ and CD4+/CD8+ in peripheral blood of RAU rats,it may be released with its immune function. 5,LBP could improve thymus quality and thymus index in RAU rats and improved its immunity.
