Rhubarb is a commonly used traditional Chinese medicine, it is cold, bitter, and with diarrhea hot bowels, stasis pass through, cooling blood detoxification effect. "Chinese Pharmacopoeia 2010 edition", defined Rhubarb as Polygonaceae Rheum palmatum L., Rheum Rheum tanguticum Maxim.ex Balf. or medicinal rhubarb Rheum officinale Baill.'s dried root and rhizome, control the quality by micro-identification, thin layer chromatography, inspection and determine the content of anthraquinones. Rhubarb is a famous medicine in Gausu Province, at present, it si still lack of spacificaty research and evaluation, and rarely report in security. This research paper in the original standards, improving the standard for overall control of the quality of rhubarb,and providing an experimental basis, technical approaches, and useful reference.The study includes the following aspects:1 Discuss TLC methods of Rhubarb:search the "Chinese Pharmacopoeia"and national standards contained Rhubarb preparation who involved reference drug and reference substances, more than 110 proprietary Chinese medicines need reference drug distinguish and 11 need reference substance. According to different extraction methods, solvents and expand systems, selecting 12 Chinese preparation medicines for TLC, the result is 8 conditions and methods are reasonable, and 4 are unreasonable which needs further study; the extrac method and expand system of TLC of Niuhuangjiedu are best,the spots were clear and better separation.2 Improve testing method of rhaponticin:Experimental techniques of paper chromatography is similar to TLC,but the solvent requir for a long time and the operation is complex. This article improved the paper chromatography, through thin layer chromatography and HPLC, increasing accuracy and sensitivity.3 To establish a method for determing of gallic acid and catechin in Rhubarb. The samples were analyzed on Waters C18 column under gradient elution. The mobile phase was methanol-0.1% ortho-phosphoric acid and the flow rate was 1.0 mL·min-1. The wavelength of UV detector was 280nm, the column temperature was room temeperature, and the quantification was performed with external standard method. RESULTS:The linear range of gallic acid was 0.106~2.125μg, the average recovery was 99.3%, and the RSD was 0.71%. The linear range of catechin was 0.465~9.296μg, the average recovery was 98.9%, and the RSD was 2.06%.4 Principal Component Analysis:determining the content of the five anthraquinones by HPLC, and using eatablished method to determin the content of gallic acid and catechin,and with the seven contents as indicators,established comprehensive evaluation model for principal component analysis for different areas of Rhubarb.concluded that the evaluation of rhubarb should be added to the control of the content of gallic acid and catechin.5 To establish the fingerprint of Rhubarb, providing good reference to the quality control of Rhubarb. the chromatographic conditions as follow:Waters C18(5μm,4.6 mm x 250 mm), The mobile phase was methanol-0.1% ortho-phosphoric acid and the flow rate was 1.0 mL·min-1. The wavelength of UV detector was 254nm. Established a total fingerprint patterns, and compared the similarity of the other origins Rhubarb, showed that it is obvious different in different aera of Rhubarb; did a cluster analysis with similarity as a means of measurement, the result were consisteng with the similarity.6 Rhubarb planting herbs in soil and heavy metal residues:by determining 5 kinds of harmful heavy metal, master the soil background, provide the basis select evidence of cultivation; and understand the effect of harmful heavy metal to rhubarb, it is necessary to limit harmful heavy metal for Rhubarb come standard quality control.
