Objective:To investigate the influence of CO2-insufflation with different pressure and different exposed time on the expression of chemokine receptor in colorectal carcinomas cell.Methods:With an artificial pneumoperitoneum model in vitro, SW480 colon carcinoma cells were exposed to CO2-insufflation at different pressure ( 6, 9, 12 and 15 mmHg ) for 1, 2 and 4 hours. The without CO2-insufflation groups were exposed to 37℃,5%CO2.The protein level of chemokine receptor CXCR4 and CCR7 were measured 0, 24, 48 and 72 hours ( culture of 37℃,5%CO2 ) after CO2-insufflation using immunocytochemistry and the mRNA level of these chemokine receptors were detected by RT-PCR.Results:In protein level the expression of CXCR4 on SW480 cell line decreased significantly in 6,9,12,15mmHg CO2-insufflation group in the same exposed time;the expression of CCR7 on SW480 cell line decreased significantly in 12,15mmHg CO2-insufflation group(P<0.05),and the expression of these chemokine receptors returned to the without CO2-insufflation level by 48,72h(P > 0.05). With the CO2-insufflation pressure ascending , the expression of CXCR4,CCR7 was decreasing in the same exposed time;with the CO2-insufflation exposed time prolonging,the expression of CXCR4,CCR7 were not significantly different in the same pressure. In the same exposed time,the transcription level the expression of CXCR4,CCR7 on SW480 cell line decreased significantly in 6,9,12,15mmHg CO2-insufflation group(P<0.05),and the expression of these chemokine receptors returned to the without CO2-insufflation level by 48h(P > 0.05). With the CO2-insufflation pressure ascending ( in the same exposed time ) and the CO2-insufflation exposed time prolonging ( in the same pressure ),the differences between the pressure groups were not significant.Conclusion:The expression of these chemokine receptors was affected temporarily after the continuous CO2-insufflation. The higher pressure of CO2-insufflation plays an important role in inhibiting the expression of these chemokine receptors.The expression of these chemokine receptors was not significantly affected after different exposed time.
