[Objective]:Colon carcinoma is one of our country most common digestive tract malignant tumors. In recent decades, with the improvement of people's living standard and changes in diet,the incidence of colon carcinoma incaeased. If chemokine and chemokine receptors are combinated, they can promote the proliferation of tumor cells,the form of the tumor vessels and the locate migration of tumor cells. Therefore, the certain indicators that can influence and monitor rectal cancer lymph node metastasis become the present research hot spots. This experiment through examines the expression of chemokine receptor CCR7 and CXCR4 in human colon carcinoma and their significance to assess their expression with the metastasis and prognosis of colon carcinoma.the combined examination of CCR7 and CXCR4 may be useful in predicting lymph node metastasis of colon carcinoma. Also the expression of CCR7 and CXCR4 could be helpful in prognosis and chems after surgery of patients with colon carcinoma.Chemokine receptor is a kind of chemokine-mediated function in the exercise of GTP-connected dual transmembrane protein receptor (GPCR), usually expressed in immune cells, endothelial cells, such as cell membrane. Elements from about 330 amino acid composition.7 transmembrane region will be divided into elements of the extracellular N-terminal free, three of the outer ring, the inner ring three cells and C-terminal parts. Chemokines by the classification will be the same type CC chemokine receptor known as the combination of CC receptors (CCR), the same category CXC chemokine receptor known as the combination of CXC receptors (CXCR), the same C and CX3C receptor (CR, CX3CR). Chemokine and chemokine receptors induced by the interaction of target cell chemotaxis migration and the rearrangement of the cytoskeleton, enhanced target cells and endothelial cell adhesion, and so on. Extensively involved in the cell growth, development, differentiation, apoptosis, and other physiological functions, and in a variety of pathological processes play an important role, such as:tumor metastasis.This experiment through examines the expression of chemokine receptor CCR7 and CXCR4 in human colon carcinoma and their significance to assess their expression with the metastasis and prognosis of colon carcinoma.the examination of CCR7 and CXCR4 may be useful in predicting lymph node metastasis of colon carcinoma. Also the expression of CCR7 and CXCR4 could be helpful in prognosis and chems after surgery of patients with colon carcinoma.[Methods]:Immunohistochemistry was used to detect the expression of chemokine receptor CCR7 and CXCR4 in 110 patients with colon carcinoma.The relationship between the CCR7 and CXCR4 expression and the clinic pathological characters was statistically analyzed.[Results]:CCR7 and CXCR4 expression were positively expressed in 59.1% and 45.5% of the patients.The high expression rate of CCR7 and CXCR4 in lymph node metastasic positive cases were 81.5% and 60.0% respectively.,but the without lymph node metastasic positive cases were 26.7% and 24.4% respectively.The positive expression was significancely higher in the patientswith lymph node metastasis than that without lymph node metastasis (P<0.05), and was also correlated with depth of invasion (P<0.05) and TNM stage (P<0.05), but not with age at surgery, gender, tumor size, histological type or different degree. In the retrospective study, the sensitivity,specificity, positive predictive value (PPV), negatively predictive value (NPV) of CCR7 express for lymph node metastasiswas 90.8%.86.7%,90.8%,86.7% respectively.the sensitivity,specificity, positive predictive value (PPV), negatively predictive value (NPV) of CXCR4 express for lymph node metastasiswas 69.2%,88.9%,90.0%,66.7% respectively.[Conclusion:]:CCR7 and CXCR4 expression were positively expressed in colon cancer.The expression of CCR7 and CXCR4 may be related to lymph node metastasis of colon cancer. Moreover, the examination of CCR7 and CXCR4 may be useful in predicting lymph node metastasis of colon carcinoma. Also the expression of CCR7 and CXCR4 could be helpful in prognosis and chems after surgery of patients with colon carcinoma.
