| Purpose: To investigate the effect factors on differentiate epidermal stem cells into neuron-like cells .Methods:The epidermal stem cells were selected and cultured by rapid attachment to a substrate, then they are identified by k15,β1-integrin,nestin and CD34. K-SFM,NEUROBASALTM-A or DMEM were added to the isolated epidermal stem cells respectively .BFGF in different concentration were added into epidermal stem cells ,morphological changes of The epidermal stem cells were observed .20ng/ml bFGF were added to different concentration epidermal stem cells in K-SFM .The changes of morphological and CD34,β1-integrin,nestin,NSE were detected.Results:1 The skin stem cells were successfully separated and cultured. They expressed characteristic skin stem cell marker K15 andβintegrin. 2.Cells of control group had a dense density.They were round or polygon. the cells were found to have high N/C ratios and the dividing cells can be found. The morphology was clear,and the cytoplasm was well-distributed. 3 In contrast, the skin stem cells in experimental group, some cell showed a bipolar morphology, These cell were founded surrounding the skin stem cells . They expressed characteristic neuron marker nestin and NSE.Conclusion:1. The epidermal stem cells were successfully separated and cultured by Using rapid adhesion method,meanwhile they expressed characteristic epidermal stem cell marker k15 andβ1-integrin.2 The epidermal stem cells were cultured in K-SFM.3. The bFGF can differentiate epidermal stem cells into neuron-like cells. they expressed characteristic neuron marker nestin and NSE. The epidermal stem cells can be differentiated into neuron depending on the cell's concentration. |
