Background: To repair the wounds caused by burns, trauma and surgery need a large number of autograft-skin and the donorsite of autograft-skin is scarcity. It is a good method to use the substitute of autograft-skin (tissue engineering skin) . The merits of a ideal tissue engineering skin ,which is permanent, sould be composed of epidermis, dermis, sweat glands, hair follicles , sebaceous glands, melanin cell and langerhans cell. Up to the present, there was no any one ideal tissue engineering skin. The important roles of tissue engineering skin are seeding cell and scaffolds. Epidermis stem cell has potential of multi-differentiation, besides multilayre epidermis, which can form sweat glands, hair follicles , sebaceous glands and so on. But it is difficulty to got the Epidermis stem cells from epidermis tissues and can not use as a seedind cell for tissue engineering skin . To explore the feasibility of differentiation of bone marrow mesenchymal stem cell (BMSCs) into epidermal stem cells, BMSCs were isolated, purified, cultrued and induced by medium-liquid of HaCaT Cell and EGF in vitro. To investigate the ability of differentiation of BMSCs into epidermal stem cells in vivo and the effects healing rabbit skin wounds, the BMSCs were labeled with BrdU(5- bromodeoxy - uridine) ,than grafted on to rabbit skin wounds. To observe the capability of scaffolds, the BMSCs were growed into Gelatin/PCL composite nanofibrous scaffolds by electrospun.
