| Objectives Through the establishment of the rat asthma model, we took montelukast sodium as the interventions, so as to comparing asthmatic rats lung, spleen and thymus lymphocytes NF-κB mRNA, lung and spleen's pathology changes in each group, thus they provides the theory basis for montelukast sodium in the prevention and treatment of bronchial asthma.Methods 36 rats were randomly divided into normal control group (Group A), asthma group (Group B), montelukast sodium group (Group C), each group has 12 mice. On the 1st and 7th day of experiments, Group A were injected into abdominal cavity with sensitization fluid (aluminum hydroxide dissolved in saline). On the 14th day of the experiments, the rats had been inhaled physiological saline (NS) for 2 weeks, daily one time, 60 minutes each time. Group B and Group C were injected into abdominal cavity with sensitization fluid of 1.0 ml / only / times (the mixtures of 0.08% OVA and Aluminum hydroxide). On the 14th day of the experiments, the rats had been inhaled 1% OVA solution for 1 weeks in order to establish asthma model, daily one time, 60 minutes each time. When the model is successful, then continue to spray for 7 days. On the 8th day after atomization inspired,Group B gives physiological saline (NS) to fill the stomach before each time OVA atomizes at the beginning of the day before OVA aerosol; Group C gives montelukast sodium (6mg/kg, NS solution) to fill the stomach before each time OVA atomizes at the beginning of the day before OVA aerosol. The time of intervention is a week. Montelukast sodium group of lung tissue, thymus, spleen lymphocytes decreased activation of NF-κB, NF-κB positive cells decreased compared with the asthma group were significantly different (P <0.05). (2) Lung tissue and spleen pathological examination: Normal control group: the bronchial lung tissue is normal, all levels of bronchial epithelial integrity, the alveolar septum is normal and the inflammatory cell infiltration is few. The spleen red marrow and white marrow structure is normal, the white marrow lymph area proliferation is not obvious. Asthma group: the number of goblet cells in lung airway epithelial cells increased, the infiltration of Submucosal lymphocytes increased, the infiltration of interstitial lymphocytes and Eosinophils was morderate and severe, alveolar septal was widened significantly, the red blood cells and lymphocytes were exudated into the alveolar space. The spleen red marrow lymphocyte is sparse, the white marrow expands, white marrow lymphocyte area moderate proliferation. Montelukast sodium group: the number of goblet cells in lung airway epithelial cells decreased, the infiltration of Submucosal lymphocytes decreased, the infiltration of interstitial lymphocytes and Eosinophils was mild to moderate, alveolar septal was not widened significantly, the red blood cells and lymphocytes were exudated into the alveolar space. The spleen red marrow lymphocyte is sparse, the white marrow mild expansion, white marrow lymphocyte area light moderate proliferation.Conclusions (1) Montelukast sodium suppress lung tissue, thymus tissue, spleen lymphocytes in the activation of NF-κB, NF-κB positive cells decreased. Montelukast sodium have a certain anti-inflammatory effect in the pathogenesis of bronchial asthma. (2) Montelukast sodium reduce the infiltration of Submucosal lymphocytes, decrease the infiltration of interstitial lymphocytes and Eosinophils, mitigate the white marrow lymphocyte area light proliferation in spleen. Montelukast sodium have a certain anti-in flammatory effect in the pathogenesis of bronchial asthma. |
