Objective: To investigate the expressions of Slit2/Robo1 in human rectal carcinomas and its correlation with micro- vascular density, biological behaviors and angiogenesis. Expect to bring some enlightenment to the cancer targeted therapy from molecular biology.Method: Select the First Affiliated Hospital of Anhui Medical University in 2009 -2010 years between surgery and pathologically confirmed in 65 cases of rectal cancer specimens as the experimental group,all patients were not receiving chemotherapy or radiotherapy before surgery,and a complete follow-up data. The other 43 patients taking the corresponding adjacent normal tissues as control group. The expression of Slit2 protein, Robo1 protein and CD105-labeled micro- vascular density ( MVD ) were measured by immunohistochemical staining (SP) in 65 cases of rectal carcinomas and 45 cases of control tissues.The use of statistical methods for analysis of the data.Result: 1. The positive rates of Slit2/Robo1 were 64.6% and 78.5% in rectal carcinomas, 4.7% and 7.0% in control tissues, and the expression of Slit2/Robo1 and MVD in cancerous tissues were higher than those control tissues (P<0.01).2. The expression rates of Slit2 and Robo1 were significantly associated with tumor size, invasion depth, Dukes stage, lymph node and distant metastasis. Statistically significant difference test.3. The MVD in the tumor tissue with positive expression of Slit2 and Robo1 was much higher than that in the negative one (P< 0.01).4. There was a significant positive correlation between Slit2 and Robo1 expression (r=0.551, P< 0.01)., Conclusion: 1. Slit2/Robo1 signaling system and the enumerations of MVD were distinctively over-expressed in the rectal carcinomas. The relation between them in cancerous tissues was positive. Speculated Slit2/Robo1 signaling system plays a major role in the formation, which may be in the process of rectal cancer angiogenesis plays a facilitating role.2. The expression of the Slit2/Robo1 and the enumerations of MVD had significant relations with the tumors, clinical features, such as invasion and lymph node metastasis. The Slit2/Robo1 signaling system and angiogenesis plays an important role in cancer invasion and metastasis.3. The expression intensity of Slit2/Robo1 and the enumerations of MVD can be used an important index for the capacity of growth, invasion and metastasis in the rectal carcinomas4. Slit2/Robo1 proteins were distinctively over-expressed in the rectal carcinomas and promote tumor angiogenesis in some way. Through the inhibition of tumors, angiogenesis by molecular targeting, could effectively control tumors, recurrence and metastasis.
