ObjectiveTo investigate the cxpression the gene of Slit2 and Robo1 in gastric cancer and gastric paracancerous tissues.To investigate the expression of Slit2 and Robo1 in both mRNA and protein level,so that to analyze the relationship among Slit2/ Robo1 pathway in human gastric carcinomas,and the relationship between Slit2/ Robo1 pathway and tumor Angiogenesis,so that we can found a new pass in diagnosis and treatment in gastric cancer.MethodRT-PCR was used to detect the transcriptase of Slit2 mRNA in 54 pairs of gastric cancer tissues and paracancerous tissues.The expression of Slit2 and Robo1 protein were respectively evaluated by immunohistochemistry,microvessel density(MVD) marker by CD34.Results1.The positive expression rate of Slit2 mRNA expressed in gastric specimens and in paracancerous tissues were70.4%and 61.1%.contrastingβ-actin the level of Slit2 were 0.61±0.21 vs 0.84±0.07,There were no significantly different with them (P>0.05).The level of Slit2 mRNA was associated with ages only in clinical characters.2.The positive expression rate of Slit2 in gastric cancer 60.7%(17/28)was remarkably higher than that gastric paracancerous tissues 21.4%(6/28)(x~2=8.9275, P<0.01). 3.The positive expression rate of Robo1 in gastric cancer 75%(21/28)was remarkably higher than that gastric paracancerous tissues 28.6%(8/28)(x~2= 12.0868,P<0.01).4.In the case of gastric cancer,There was a positive correlation between expression of slit2 and Robo1,(Pearson r=0.4202,P<0.01).The expression of slit2 was remarkably associated with tumor volume,clinical stage,(P<0.05);the expression of Robo1 was remarkably associated with tumor volume,clinical stage and lymph-node metastasis,(P<0.05).5.The MVD of the slit2 positive expression group(29.64±3.11)was remarkably higher than that of the negative expression group(20.41±1.73),so as the expression of Robo1(27.91±2.40 vs 20.42±1.08),otherwise,the MVD in cancer group was higher contrast normal tissues.Conclusions1.In gastric cancer,Slit2 are not constitutively expressed in mRNA level,the disorder expression may occur in translation level.It proves that the generation of gastric cancer was a multi procedure and multigene process.2.There was a positive correlation between expression of slit2 and Robo1,but the expression were not parallel.3.Slit2,Robo1 and MVD made tumor cells more invasive,their over expression may lead to advanced invasion of gastric cancer and unfavorable prognosis.slit2 may encourage tumor angiogenesis by effect Robo1.MVD correlated with Slit2 and Robo1 may attribution to result of angiogenesis adopt the rapid grow of gastric cancer.SenseThe efforts of nerve migration factor slit2 in generation of gastric cancer and tumor angiogenesis prompt that between vascular system and nerve system may have similar mechanisms,It might have an important sense of the tumer diagnose and anti angiogenesis therapy.
