Objective To investigate the influence and molecular mechanism of RNA interference targeting P115 on the invasion of human gastric cancer BGC-823 cells.Methods The recombinant of pGPU6/GFP/Neo/P115-1318 was transfected into BGC-823 cells with Lipofactamine 2000.Fluorescence microscopy was used to observe and calculate the efficiency of transfection,P115 protein and mRNA expression were detected by Western-blot and RT-PCR to estimate braking effect ;MIF,MMP2,MMP9 protein expression were detected by Western-blot and immunofluorescence method;RT-PCR was adopted to detect mRNA expression;Wound closure assay and Transwell chamber assay were performed to detect the invasion ability of BGC-823 cells.Results pGPU6/GFP/Neo/P115-1318 was stably transfected into gastric cancer BGC-823 cells,48 hours later,the average efficiency of transfection was 68%;To compared with other two control groups(blank control group and negative control group) ,P115,MIF,MMP2,MMP9 protein and mRNA expression of the recombinant of pGPU6/GFP/Neo/P115-1318 were obversely reduced (P<0.05);Wound closure assay and Transwell chamber assay showed that the number of cell invision in P115shRNA group was significantly fewer than the control groups(P<0.05).Conclusion Interference suppression of P115 may inhibt invision of BGC-823 cells by down regulating the expression of MIF,MMP2 and MMP9 .
