[Object]To determine the expression level of activating transcription factor 6 (ATF6) and double-stranded RNA-activated protein kinase-like endoplasmic reticulum kinase (PERK) in the brain tissues from patients with refractory temporal lobe epilepsy and explore their potential roles in epileptogenesis.[Methods]30 patients who underwent surgical treatment for intractable temporal lobe epilepsy and chosen in the study,as well 10 age-and gender-matched controls with cerebral operations for decompression or debridement.All cortical tissue was obtained quickly in ice box after resection.The process was bioclean. Immunohistochemistry was used to examine the expression of activating transcription factor 6 (ATF6) and double-stranded RNA-activated protein kinase-like endoplasmic reticulum kinase (PERK).[Results]Gliosis was observed, to different extent, in the brain tissues from patients with refractory temporal lobe epilepsy,so were neuronophagia phenomenon and neuronal degeneration. The expressions of ATF6 and PERK protein were significantly higher in the temporal lobe of patients with refractory temporal lobe epilepsy than the controls(P<0.05).[Conclusion]The expressions of ATF6 and PERK were upregulated in the temporal lobes of patients with refractory temporal lobe epilepsy,which suggests their potential roles in epileptogenesis.
