Purpose:Epilepsy is a chronic neurological disease. Studies have shown that repeated seizures or status epileptics can lead to damage of the brain tissue, especially hippocampus, and brain damage can be caused by seizures. It has been widely studied the relationship between the epileptogenesis and the hippocampus damage, but there is only a little research on the correlation about the axon guidance factors Eph / Ephrin and epileptogenesis. In this experiment, it is observed that the role of the Eph / Ephrin in the axon reconstruction and synaptic plasticity after epilepsy. It will provide a theoretical basis for the further study the pathogenesis of temporal lobe epilepsy and clinical treatment.Method: Status epilepticus mouse model is induced by pilocarpine administered. They are observed the dynamic expression of EphA / EphrinA and mossy fiber sprouting (MFS) in mice's hippocampus.Result:1.RT-PCR method found the expression of EphA/EphrinA mRNA, EphA4(p<0.01),EphA8(p<0.05),EphA10(p<0.05),EphrinA2(p<0.05), EphrinA3(p<0.01) and EphrinA4(p <0.05) have generally increased at the day 7 after status epilepticus comparing with the control groups.2. Comparing with the control groups, the expression of EphrinA4 mRNA has an increase in the CA3/ CA1 area of hippocampus after epilepsy using in situ hybridization dec, which is expressed the peak at the day 7 (P <0.05) in the CA3 area and at the day 14(P <0.05) in the CA1 area , generally. But there was no significantly difference in the DG. The expression level of EphA10 mRNA is significantly increased at the day 7(P <0.05) after epilepsy, and up to peak at the day 14 (P <0.01) in the CA3 and at the day 7 (P <0.01) in the CA1 area. There is the significant increase at the day 14 (P <0.05) in the DG area.3. By immunofluorescence histochemistry technique, we find that EphA10 protein expression has the increased at the day 3 and up to peak at the day14 in the CA1 and CA3 area (P <0.01) but not in the DG. 4. It is not appeared mossy fiber sprouting both the natural and epileptic mice in inner molecular layer of DG by Neo-Timm staining, but it begin to appear from day 7 in the CA3 areas and increased gradully from the day 7 to the day 28 ,from CA3a area to CA3c area.Conclusion:1. Semi-quantitative RT-PCR technology is used to research the expression of EphA / EphrinA mRNA in the hippocampus, the results showed that the expression level of these molecules occurred varying degrees increase after status epilepticus. It's implied that they may be involved neurons loop reconstruction in hippocampal.2. By in situ hybridization method, it is confirmed that EphrinA4 and EphaA10 mRNA distribute widely in the hippocampus and increase after status epilepticus. So, we consider that EphrinE4 and EphA10 may be involved axonal sprouting phenomenon in the hippocampal.3. Immunofluorescence histochemistry method is used to detect the distriction of EphaA10 protein in the hippocampus. The result showed that expression level of EphaA10 protein increased at first and decreased then during epilepsy. It is suggested that EphA10 may be associated with the mossy fiber sprouting following chronic epileptogenesis .
