Objectives:To observe the effects of polysaccharide from the Uygur Herb Hyssopus officinalis L.on the hypothalamic-pituitary-adrenal (HPA) axis, cytokine, the function in the morbility of asthma and the correlation between each other. Discuss parts of mechanisms on its treatment of asthma and provide experimental basis to clinical application. Methods:(1) the optional extraction process of polysaccharide from Hyssopus officinalis L. was selected with the orthogonal design. (2)Rats were randomly divided into control group, asthmatic group, Dexamethasone group, low-dose of polysaccharide from Hyssopus officinalis L. group and high-dose of polysaccharide from Hyssopus officinalis L. group. The rats were sensitized with association and then challenged with repeated inhalation for preparation of asthmatic model. All specimens were adopted from anesthesia rats in 2 hours after last OVA aerosol inhalation. The levels of IL-4, IFN-γ, IL-17 and CORT in serum were detected with ELISA, the expression of CRH mRNA was detected with RT-PCR, and the expression and location of IL-6 in lungs were analyzed by Immunohistochemical. Results:(1) The optimum extraction process was as follows the polysaccharide was extracted with for 1 hour with 25 times water for 2 times and alcohol concentration was at 85%. (2) Compared with control group, the levels of IL-4, IL-17 in serum and IL-6 in lungs heightened significantly (P<0.05), the levels of IFN-y, CORT in serum and the expression of CRH mRNA in hypothalamus was declined significantly in those of asthmatic group (P<0.05). (3) Compared with asthmatic group, the levels of IL-4 and IL-17 in serum and IL-6 in lungs were significantly lower (P<0.05), the levels of IFN-γ, CORT in serum and the expression of CRH mRNA in hypothalamus was heightened significantly in those of treated groups.(4) Compared with dexamethasone group, there were some differences between the levels of IL-4, IL-17, IFN-γ, CORT in serum, IL-6 in lungs and CRH mRNA in hypothalamus in low-dose of polysaccharide from Hyssopus officinalis L group (P<0.05), and no differences in high-dose of polysaccharide from Hyssopus officinalis L group except IL-6 in lung. (5) Compared with low-dose of polysaccharide from Hyssopus officinalis L group, the levels of IL-4 and IL-17 in serum and IL-6 in lungs decreased significantly(P<0.05), the levels of IFN-γ, CORT in serum and the expression of CRH mRNA in hypothalamus were heightened in those of high-dose of polysaccharide from Hyssopus officinalis L group. (6) the levels of IL-4 had negative correlation with IFN-γin serum (r was-0.408, P<0.01); there is positive correlation between inflammatory cytokines IL-4 with IL-17 and IL-6, IL-17 with IL-6 (r was 0.542,0.561 and 0.694, respectively, P<0.01); The levels of inflammatory cytokines (IL-4, IL-6, IL-17) had negative correlation with CORT in serum (P<0.01), r was -0.428,-0.533 and -0.697, respectively. (7) IL-6 observed in bronchial epithelium cells, inflammatory cells and parts of interstitial lung cells, while in hyalomitome, IL-6 reached peak in asthmatic group, the level in treatment groups are significantly drops(P<0.05). Conclusions:The function of HPA axis was inhibited in asthma models, nerve endocrine hormone secreted decrease and accompanied with the increase of the production of IL-4, IL-6 and IL-17, the chronic inflammation in asthma aggravated; Polysaccharides from Hyssopus officinalis L. could adjust function of HPA axis in asthma, improve the endogenous anti-inflammatory mechanism, still can inhibit the release of inflammatory cytokine and improve the inflammation progress in asthma.
