Prostate cancer patients accounted for 11% of all cancer patients, and of the cancer deaths 9%. In China, prostate cancer rates are lower than in the western countries, but in recent years has gradually rising trend. The cause of prostate cancer has not been clear. But the genetic factor is already confirmed one of the most important factors.Prostate cancer patients can not have any symptoms, the diagnosis depend mainly on a digital rectal examination, PSA, the rectum b-sonography, prostate biopsy and imaging findings. All its various check have certain defects, in the early diagnosis may have certain difficulties. Miss the optimal period of treatment, poor outcome, patients' pain.Recent research of E2F3 gene is very common. Its expression has certain relationship with prostate cancer about the degree of malignancy. In the occurrence and developing of prostate cancer it has stimulate influence, E2F3 gene plays a central role in cell cycle regulation as transcription factor. E2F3 participate in the cell cyole control through the interaction of pRb and related proteins, such as the cell cycle dependent proteins and kinase, plays an important regulation role in the cell cycle and DNA replication, it controls the process of cell proliferation and many genes expression. It is a necessary transcription factor during cell synthesis by G1 into S period midterm. E2F3 and pRb have a close relationship. The main function of the pRb is growth inhibition through the adjustment of E2F3. The high expression of E2F3 often indicates poor prognosis. Detection of expression of E2F3 helps the judgment of prostate cancer treatment and malignant degree and prognosis. E2F3 is expected to become a diagnosis of prostate cancer and evaluation index of prognosis.RNAi is a common prevalent conservative phenomenon in the biological evolution procession. It is motivated by small, double-stranded RNA, based on the degradation of the target genes mRNA caused gene transcription silence. RNAi is a very conservative gene silencing mechanism, in primitive organism RNAi can protect genome from inserting viruses or other gene fragments, and adjustment numerous gene express in the biological development process. AAV carrier is the most extensive research in recent years, the biggest concern gene therapy vectors, with wide host range, low immunogenicity and high security, stability. AAV carrier is considered as the most promising gene therapy of the new generation virus carrier Objective:Explore the efficiency of AAV carrying PC3 cells. Analysis the relationship among E2F3, AR and HIF-1αthrough testing the expression of the three gene in PC3, aiming to explore the relationships of E2F3, AR and HIF-1αin prostate cancer, and compare the effects of E2F3, AR and HIF-1αin cell proliferation.Methods:Cultivating cells of PC3 in vitro, explore the best condition of cell growth. AAV virus carrying cultivated PC3, capture pictures of AAV virus carrying cultivated PC3. And calculate carrying rate to explore the best carrying concentration and time. Explore the growth activity of AAV carrying PC3 cells with MTT method, rendering the growth curve. Research E2F3, AR and hypoxia-inducible PC3 cells-1 alpha in the mutual relationship in expressionResults:AAV can highly carry PC3. After AAV roling with PC3 60 minutes, Use fluorescent microscopy can observe green fluoresce expression after 24hr, the optimal concentration of AAV carrying PC3 is 1×105v.g/cell. In 24h,48h and 72h fluorescence have trend of enhancement of expression (P< 0.05). Determined by MTT method detection, the higher the concentration of AAV the grow activity of PC3 cells decline (P<0.05). Thermo Fluoroskan Ascent method can detect fluorescent protein expression of E2F3, AR and HIF-1αin PC3. There is a certain relationship between the three.Conclusion:AAV can carry PC3 effectively, and have influence on its growth. The expression of E2F3, AR, HIF-1αare certain correlation. This experiment can provide reference for follow-up tests.
