ObjectiveTo explore the influence of trichosanthin on the proliferation of ovarian cancer HO8910, trichosanthin-induced apoptosis in ovarian cancer HO8910 cells, and apoptosis relation whit genes Bax, Bcl-2 expression mechanism.MethodsIN this study ovarian cancer HO8910 cells(logarithmic growth of the cells).Experimental groups for the control group, experimental group: TCS20,40,80μmol/L. The inhibitory proliferation effect of TCS on ovarian cancer HO8910 cells was detected by MTT.Cellular cycle phase was measured with flow cytometry.The expressions of Bax, Bcl-2 genes were detected by Western Blot.ResultsCharacterrstic changes of cell apoptosis such as volume of cell minishing and protoplasm constricting were measured and subdiploid peak can be analysed,the gene expression of Bcl-2 decreased but the gene expression of Bax increased along with trichosanthin,s concentration.ConclusionTrichosanthin can irestrain the growth of ovarian cancer HO8910 cells,and induce apoptosis in ovarian cancer HO8910 cells ,the down regulation of Bcl-2 gene and up regulation of Bax genemay be the mechanisms of the apoptosis expression .
