Objective To study the the relativity of galectin-7 and acute rejection and to explore the role of galectin-7 in propression of allograft rejection.Methods Eighteen male mice were randomly allocated into two groups: surgical control group,acute rejection experimental group. The surgical control groups had syngeneic mice skin transplant ( C57BL/6→C57BL/6 ) ,while the acute rejection experimental groups had allogeneic mice skin transplan(tBALB/c→C57BL/6).The animals were sacrificed at 10 days after operation under sterile circumstance. The existence of differences of galectin-7 protein expression level of the graft between the two groups were analysed qualltatively by using immunohistochemical SP method. Meanwhile the differences of the protein expressions of galectin-7 between the two groups were determined by western blot. Mean comparison in groups was conducted with the two independent-samples t test.Results Frozen section and Immunohistochemistry staining showed that the expression of galectin-7 of graft was positive in the surgical control group and nagetive in the acute rejection experimental group. Expression of galectin-7 of graft was positive both in surgical control group and acute rejection experimental group,interestingly, the expression level in the acute rejection experimental group was significantly lower than in the surgical control group, showed by Western blot. The difference of the expression of galectin-7 between the two groups was extremely significant(t =4.476, P<0.05).Conclusion The expression level of galectin-7 in the graft decreased in acute rejection after allogeneic mice skin transplant. Combined with The inhibition function of the other members of the galectin family on lymphocyte proliferation, we speculated that the apoptosis-promoting effects or the inhibition proliferation function of galectin-7 protein on T cell which played a major role in acute rejection reduce the number of T cells, which restrained acute rejection.so galectin-7 may be one of inhibitory factors of acute rejection.Meanwhile,the protein was expected to be a marker for diagnosing acute rejection in clinical practice and a new thought for the treatment of cute acute rejection.
