Chicken anemia virus is a special kind of circinal sigal DNA virus,which code three kinds proteins: VP1,VP2 and VP3. VP3(Apoptin) has shown to induce apoptosis in various tumor cells, but not in normal human cells and murine diploid cells. So VP3 has a very promising on anti-tumor biological medical treatment. But the specific tumor cell apoptosis mechanism is not clear, the research has shown VP3 protein in cell line may be closely related with apoptosis. It is well known that the alterations of the protein primary structure can affect the specific biological functions of the protein, for the conformation is the foundation of the nature and functions of proteins. In the experiment, we aim to investigate the soluble protein expression as well as the activity of the protein after the removal of the meaningless apoptin-recombinant gene. We use the PCR to get rid of the useless sequences of the HIS-PTD4-VP3 gene, and get two recombinants. Then we detect the solubility of the three kinds ofVP3 proteins by PAGE; detect the biology function by FCM, to compare the capability of inducing the apoptosis of the tumor cells. The result demonstrated that structural change of VP3 protein can affect its solubility and the biological function. We can draw the conclusion from the experiment that the solubility and the activity of the protein that expressed by recombinant 1 gene has been elevated.
